Serum Pharmacochemistry Of Gentianella Acute And Pharmacokinetics Of DMB

Gentianella acuta has long been the Mongolian medicine for treatment of jaundice hepatitis、gastritis、fever、headache and so on.In some areas are used to cure heart disease, local known as the "steady heart grass".Gentianella acuta contains a lot of xanthones. Researchers have proved that swertianolin and bellidifolin of Gentianella acuta can ease the heart rate too fast.Gentianella acuta has not established a standard quality standard.Object: Simultaneou determination of Swertianolin, bellidifolin, desmethybellidifolin of Gentianella acuta byHPLC.Methods:The analysis was aehieved on Waters Symmetry Cig column(4.6mm× 250mm,5μm) using a mobile phase of MeOH-Water(pH2.6) gradient elution in a total run time of 45 minuetsand the detection wavelength was set at 252nm. Results:Swertianolin,desme-thybellidifolin and bellidifolin were linear over the range of 13.6～122.4μg/ml,10-90μg/ml,10.8-97.2μg/ml. The average recoveries (n=6) were 99.97%,102.02%,98.00%.The content of swertianolin, desmethyl-bellidifolin and bellifolin in Gentianella acuta were 23.94mg/g,4.49mg/g,7.92mg/g.Studying on the Gentianella acuta absorb into blood components. Gentianella acuta which absorbed into Rat’s blood had nine transitional compo-nents.The methylbelli-difolin is prototype components absorb into the blood. Conclusion:demeth-ylbellidifolin of Gentianella acuta directly in vivo.The pharmacokinetic parameters were determined after oral administration of demethylbellidifolin (28.287mg/kg) of Gentianella acuta to rats. The main pharmacokinetic parameters of demethylbellidifolin in rats were t1/2αof 0.117±0.003h, t1/2p of 12.109±4.157h, Cmax of 134.959±16.649μg/ml, Tmax of 15min.Our study established content of stable and efficient method for the determination of research. And a preliminary study on the blood components of Gentianella acuta which absorbed by rats.The pharmacokinetic of demethyb-ellidifolin provides experimental basis for the use of clinical practice.
