Objective: To investigate the expression of TGF-β1, VEGF in primary & recurrent pterygium as well as the correlation among their expression, the aim of this study was designed to test the correlation between transforming growth factor-β1(TGF-β1), vascular endothelial growth factor(VEGF) and angiogenesis in pterygium of primary and recurrent.Methods: 30 primary pterygium samples including 21 primary pterygium samples and 9 recurrent pterygium samples and 8 normal conjunctivas are collected in this study. All pterygium and normal conjunctival tissue morphological differences were showed by HE staining. Immunohistochemical Elivision staining method was adopted in detecting the expression of TGF-β1 and VEGF in cases of pterygium and normal conjunctival tissues. The results were compared between the pterygium and normal conjunctival tissues. In the pterygium group, the results were further compared between the primary and recurrent group. Statistical analysis was used to analyze the correlation between TGF-β1 and VEGF.Results: Fibroblasts proliferated actively, increased angiogenesis and inflammatory cell infiltration, were seen in the pterygium epithelium, as well as sub-epithelial tissue. There are high expression of TGF-β1 and VEGF in all parts of pterygium, and slight expression in conjunctival tissues. The expressions of TGF-β1, VEGF in pterygium were 63.33%, 70.00%, meanwhile 25.00%, 12.50% of conjunctival tissues. The expressions of TGF-β1 and VEGF in 21 cases of primary pterygium 57.14%, 88.89% and 57.14%, 100% in 9 recurrent cases. All the results of immunohistochemical staining showed the expression of TGF-β1 and VEGF in pterygium was higher than that in normal conjunctiva tissues(P<0.05). The results of immunohistochemical staining showed a significant positive correlation between TGF-β1 and VEGF in pterygium group and recurrent pterygium(P<0.05).Conclusions: The high expressions of TGF-β1 and VEGF were observed in all pterygium tissues, especially in recurrent group. TGF-β1 and VEGF present a promoting effect each other. There may be synergistic action between two factor during the development and recurrence of pterygium.
