Objective:By primary cultured human pterygium fibroblasts,and observe how the (-)Epigallocatechin gallate (EGCG) affect on HPFs proliferation,and Matrixmetallo proteinases -9 (MMP-9) and Vascular endothelium growth factor (VEGF) expression, analyze the relationship between EGCG and MMP-9 and VEGF.To provide new theoretical foundation for EGCG to be used in pterygium.To investigate new treatment of pterygium.Methods:1. Primary culture of pterygium in vitro to get fibroblasts, cells were treated with EGCG at different concentration for 72 hours, and set the control, the cells were observed under the microscope and HE staining.2. Assay EGCG inhibitis human pterygium fibroblasts'proliferation by MTT.3. Detect MMP-9 and VEGF protein expression by Western Blotting.Results:1. Cellular morphology to be change significantly, such as nuclear condensate, cell contraction and collapse by the EGCG concentration increase.Cell morphology more irregular, which was dose-dependent.2. EGCG inhibits the proliferation of HPFs obviously by time- and dose-dependent. The effect of EGCG of 80ug/ml and 72h, the inhibition rate up to 89.6%.3. With the 20ug/mland 40ug/ml EGCG after 48 hours,control to blank group, MMP-9 and VEGF protein expression levels were decreased significantlyConclusion:1. The efection of different concentrations and time of EGCG on HPFs, cells proliferation decreased, and showed a dose- and time- dependent significantly.2. The protein expression of MMP-9 and VEGF downregulated with EGCG after 48 hours,and showed dose dependent obviously.
