Objective: To investigate the clinical significance of the expression of XAF1, CHD5 and Ki67 in ovarian serous adenocarcinoma.Methods: Pathological observation of 63 cases of ovarian epithelial tumor resection, 15 cases of ovarian benign tumor, 13 cases of borderline ovarian cystadenoma,35 cases of ovarian serous adenocarcinoma. 5 cases of uterine fibroids were selected as the control group. All patients were not treated with radiotherapy, chemotherapy and hormone therapy before surgery.All cases were confirmed by operation. This study followed the seventh edition of TNM’s staging guidelines for malignant tumors. The expression changes of XAF1, CHD5 and Ki67 proteins in the tissues were detected by immunohistochemical technique, and the relationship between the expression and clinical pathological parameters and prognosis was analyzed.Results: 1 Expression of XAF1, CHD5, Ki67 protein in ovarian serous adenocarcinoma, borderline, benign tumor and normal ovarian tissueThe positive signal of XAF1 protein was mainly localized in the nucleus and / or cytoplasm, and was found in the nucleus and / or cytoplasm. The positive expression rate of XAF1 in 5 cases of normal ovarian tissues was100%(5/5). The positive expression rate of XAF1 in ovarian benign tumor,borderline tumor and ovarian cancer was 86.7%(13/15), 61.5%(8/13) and48.6%(17/35).CHD5 protein was mainly located in cytoplasm and yellow to brownish yellow granules. The positive expression rate of CHD5 protein in 5 cases of normal ovarian tissues was 100%(5/5), 93.3%(14/15), 61.5%(8/13) and12/35(34.3%), and the positive expression rate decreased gradually,comparison has statistical difference.The positive signal of Ki67 protein was mainly located in the nucleus,which was yellow to brownish yellow granules. The rate of positive expression in serous epithelial ovarian carcinoma(80%, 28 / 35) was significantly higher than that in ovarian serous cystadenoma(46.2%, 6 / 13),benign tumor(26.7%, 4 / 15) and normal ovarian tissues(20%, 1 / 5, P < 0.01),the difference was statistically significant.2 Relationship between XAF1, Ki-67, CHD5 protein expression and clinical stage, histological grade and lymph node metastasis in ovarian serous adenocarcinomaThe expression of XAF1 and CHD5 in ovarian serous adenocarcinoma was significantly correlated with clinical stage, histological grade and lymph node metastasis(P<0.05), XAF1 low expression was more common in low differentiated adenocarcinoma and III-IV stage ovarian cancer, and the positive expression rate of XAF1 protein in ovarian cancer tissues with lymph node metastasis was lower.The positive expression rate of CHD5 protein in ovarian serous adenocarcinoma was significantly negative correlated with histological grade,and the positive expression rate of CHD5 protein was lower, but there was no significant difference in clinical stage and lymph node metastasis(P> 0.05).The positive expression rate of Ki67 protein was positively correlated with histological grade, clinical stage and lymph node metastasis of ovarian serous adenocarcinoma, The positive expression rate of Ki67 protein in patients with differentiation, clinical stage and lymph node metastasis was higher.3 Survival analysisBased on age, clinical stage, pathological type and follow-up, the mean survival time and 5 year survival rate were calculated. The average survival time of 35 patients was 51.4 months, 23.5% year survival rate was 5, and the average survival time was 89.4 months, and the 77.5% year survival rate was5. The average survival time of >50 years old was 54.8 months, 5 years survival rate was 23.5%, the average survival time was 77.5 months, the66.7% year survival rate was 5. Patients with low XAF1 expression wereassociated with high differentiation of tumor and the survival and overall survival of the patients were lower than those of the control group(P=0.013).The disease-free survival and overall survival of patients with low CHD5 expression were also lower than those in the control group(<0.05 P). Cox proportional hazard regression analysis showed that XAF1 and CHD5 were independent prognostic factors of ovarian serous adenocarcinoma. Patients with low CHD5 expression, CHD5 low expression and clinical status(<0.05P), but with the age of patients, tumor type and grade, recurrence and clinical stage(P> 0.05). Survival analysis showed that XAF1 was associated with high differentiation of tumor and the survival time of patients was lower than that of control group(P=0.013). Statistical analysis of all data using SPSS15.0software package. The clinical and pathological parameters of XAF1, CHD5 and were correlated with chi square test and t test. Kaplan-Meier method, log rank test and multivariate Cox proportional hazard regression analysis of survival rate. Numerical values are represented by mean of the mean. P < 0.05 that was statistically significant.Conclusion:1 The positive expression rate of XAF1 protein in ovarian serous adenocarcinoma tissues was significantly lower than that in normal ovarian tissues, benign tumors, borderline tumors, and was negatively correlated with the degree of differentiation, clinical stage, lymph node metastasis and prognosi. Suggesting that XAF1 protein in ovarian serous cystadenocarcinoma of the occurrence and the development process has a important role, related to ovarian cancer and malignant progression.2 The low expression of CHD5 protein was associated with the occurrence of ovarian serous adenocarcinoma, and negatively correlated with the degree of tissue differentiation and prognosis, suggesting that CHD5 is a potential molecular marker and / or therapeutic target in ovarian cancer, which is expected to be a predictor of ovarian cancer survival.3 The expression of Ki67 protein in ovarian serous carcinoma was significantly increased, and it was significantly correlated with the clinicalstage of tumor, and increased with the increase of tumor malignancy, which indicated that Ki67 protein had important significance in the process of ovarian cancer.
