Objective To explore the feasibility of diagnosing acute myocardial infarction with surface-enhanced Raman spectroscopy through detecting,analyzing and comparing the spectroscopy of blood serum obtained from healthy people and the patients identified having acute myocardial infarction.Methods Patients suffering chest pain and chest tightness within 12 hours and highly suspected with acute myocardial infarction were chosen from September 2013 to March 2014 in our hospital.Among them,32 patients with clinical confirmed AMI according to the WHO guidelines were designed as the experimental group,while a total of 32 healthy volunteers were chosen as the control group.3ml peripheral venous blood were drew from each people after informed consent and blood serum was got by collecting supernatant after 3500r/min centrifugation for 10 minutes.The spectral signals obtained by detecting all of the blood serum samples with SERS technique were collected and analysed by WIRE 2.0 software package.Then the mean spectrum,prominent Raman peaks and different spectrum between two groups were obtained.Sensitivity and the specificity of the SERS detection method were obtained by employing principal component analysis and linear discriminant analysis to discriminate AMI patients from Clt subjects.Finally,receiver operating characteristic(ROC) testing is conducted to verify the effectiveness of PCA-LDA algorithm.Results The prominent SERS peaks of the two groups are at the Raman shift 494, 591, 640, 731, 813, 888, 960, 1006, 1074, 1095, 1135, 1206, 1330, 1450, 1580, and1656cm-1, respectively,and all observed SERS peaks tentative assignment is listed in Table 2.Through the comparison of the average serum SERS spectrum of the two groups found that,the signal of AMI blood serum SERS peaks shows obvious increase in the peaks at 494, 640, and 1656cm-1and decrease in the peaks at 731, 1330 and 1450cm-1.The sensitivity and the specificity of the SERS technique in the diagnosis of acute myocardial infarction were 87.5% and 93.8% by employing principal component analysis and linear discriminant analysis to discriminate AMI patients from Clt subjects.The integrated area under the ROC curve was 0.933,which proved that the effectiveness of the PCA-LDA diagnostic model we built.Conclusion The SERS technique has a high value in the diagnosis of acute myocardial infarction,and it has a certain value in clinical application.On the other hand,it can study the acute myocardial infarction at molecular level,and it is helpful for studying the mechanism after the happening of acute myocardial infarction.The method by detecting blood serum with SERS technique has a great potential to be a tool for AMI in early diagnosis and the studying of mechanism.
