In this thesis, we applied Raman spectroscopy and surface-enhanced Raman spectroscopy (SERS) techniques to preliminarily research the blood and saliva in patients with acute myocardial infarction (AMI). Using PCA-LDA multivariate statistical analysis techniques, we respectively analyzed the blood and saliva samples and compared and summarized the discriminated results of different methods to find a rapid, accurate and non-destructive method for diagnosis of AMI. The primary research as follow:1. Raman spectroscopy was applied to analyze the blood serum from AMI patients (n=32) and healthy controls (n=32). Using PCA-LDA multivariate analysis, we were able to differentiate AMI from normal with diagnostic sensitivity and specificity of 78.1%.2. Gold nanoparticle based on SERS was applied to analyze the blood serum from AMI patients (n=32) and healthy controls (n=32). Using PCA-LDA multivariate analysis, we were able to differentiate AMI from normal with diagnostic sensitivity of 75.0% and specificity of 71.9%.3. Silver nanoparticle based on SERS was applied to analyze the blood serum from AMI patients (n=31) and healthy controls (n=31). Using PCA-LDA multivariate analysis, we were able to differentiate AMI from normal with diagnostic sensitivity of 87.1% and specificity of 93.5%, which was better than that of gold nanoparticle SERS diagnosis. All the experimental results above illustrated the silver nanoparticle based on serum SERS technique for early diagnosis of AMI had a good application prospect and might become a rapid and accurate diagnosis of AMI method.4. Raman spectroscopy was applied to analyze the saliva from AMI patients (n=30) and healthy controls (n=31). Using PCA-LDA multivariate analysis, we were able to differentiate AMI from normal with diagnostic sensitivity of 80.0% and specificity of 80.6%. The experimental results showed that the Raman spectroscopy of saliva for early diagnosis of AMI had a good application prospect and might be a rapid, accurate, and non-destructive diagnosis of AMI method.
