Raman spectroscopy is a useful technology to research the molecule structure. The range of application of Raman scattering is widened with the development of various technologies. Raman spectroscopy applications to biochemistry, biophysics, biomedicine are developed fast in latest years. Structure information is available through the Raman spectroscopy of biological molecules and supply experimental basement for fast and early diagnosing of many diseases.This thesis begins with the discovery, the background of research, the developing processes and the applications of Raman scattering. Many techniques of Raman spectroscopy are introduced and surface enhanced Raman scattering is recommended in detail. Application of Raman spectroscopy in field of biomedical samples and single molecule detection are researched in this article.Firstly, Raman scattering application in biomedicine is researched. Normal rat serum, normal human serum and human melanoma cells were samples in the experiment. Contrasting the Raman spectroscopy and surface enhanced Raman spectroscopy of normal rat serum and analyzing the difference to research the enhanced effect. Most of the frequencies are signed to get the structure information of sample. Surface enhanced Raman spectroscopy of human serum is collected to research abundant vibration information of human serum and protein structures and provide experiment basement for future research. Normal Raman spectroscopy of human melanoma cell is gotten for analyzing the cell structure in molecule level, especially researching the vibration information. Most of the frequencies are signed. The experiments have shown the foreground of fast and early diagnosis of human disasters.Secondly, a novel silver sol is produced to be the substrate of surface enhanced Raman spectroscopy. Rhodamine 6G is detected in single molecule level based on this novel silver sol substrate. The substrate is proved to be useful. The reason why the strong effect of surface enhanced Raman spectroscopy is analyzed. The function of "hot spot' is highlighted in this paper.
