Objective:To evaluate the diagnostic value of surface enhanced Raman spectroscopy in laryngeal squamous cell carcinoma(LSCC)tissue and serum.Methods:From December 2016 to December 2017,a total of 39 cases tissues of laryngeal squamous cell carcinoma(LSCC),35 cases of adjacent normal tissues,35cases of serum samples preoperative and 32 cases of serum samples the 7th day after operation were collected from the Department of Otorhinolaryngology-head and neck surgery of Union Hospital of Fujian Medical University,then measured by confocal Raman spectrometer in Via.The original spectra of SERS were treated uniformly,and the spectral data were analyzed by statistical methods such as PCA-LDA?ROC curve.Results:1.The average SERS spectra of laryngeal squamous cell carcinoma tissues and adjacent normal tissues were about 400-500 cm~-11 band.The Raman intensity of SERS spectra in laryngeal squamous cell carcinoma tissues was higher than that in paracancerous tissues,but in 1400-1750 cm~-11 band.The Raman intensities of SERS spectra in laryngeal squamous cell carcinoma tissues were lower than those in paracancerous tissues,and there was no significant difference in the shape of the mean SERS spectra between the patients with laryngeal carcinoma preoperative and on the 7th day after operation.2.The discriminant model was established by PCA-LDA.The results showed that the SERS could distinguish the laryngeal squamous cell carcinoma(LSCC)tissues,the adjacent normal tissues could obtain 98.4%specificity and 99.2%sensitivity,and the preoperative serum?the 7th day after operation serum of laryngeal cancer patients could be distinguished.The specificity and sensitivity were 66.7%and 69.2%.3.After PCA-LDA multivariate statistical analysis,the accuracy and feasibility of this diagnosis algorithm in distinguishing laryngeal squamous cell carcinoma tissues from adjacent normal tissues,preoperative serum and postoperative serum on the 7th day after operation were further evaluated by using the receiver operating characteristic curve(ROC curves),The AUC values of ROC curves were 0.996 and 0.725,respectively.Conclusion:Laser surface enhanced Raman spectroscopy can be used to distinguish laryngeal squamous cell carcinoma from adjacent normal tissue.The specificity and sensitivity are 98.4 and 99.2 respectively,which provide a new nondestructive method for clinical diagnosis of laryngeal carcinoma.
