Objective:1.To investigate the expression of EZH2, VEGF and Caspase-3 in colorectal adenocarcinoma, adenoma and adjacent mucosa.2. Analyzing the relationship between all three above and clinical pathological parameters.3. Evaluating the correlation between the three.Method:Collecting 108 cases of colorectal adenocarcinoma, 40 cases of adenoma tissues, and 40 cases of adjacent mucosa excised by First Hospital of Shanxi Medical University during Jan. 2012- Dec. 2014. Testing the expression of EZH2, VEGF and Caspase-3 by IHC Polink1. Selecting fresh specimens of 30 cases of colorectal adenocarcinoma, adenoma tissues, and adjacent mucosa respectively. Testing the quantity of EZH2 protein by Western blot. Then analyzing the relationship between clinical pathological parameters and the three bio-markers above, and evaluating the correlation between the three molecular bio-markers.Result:1. In colorectal adenocarcinoma, adenoma and adjacent mucosa, expression rate of EZH2 was 80.56%(87/108), 62.50%(25/40), 5.00%(2/40), and expression rate of VEGF was 54.63%(59/108), 35.00%(14/40), 0%(0/40). Compared with corresponding tissue in adenoma and adjacent mucosa, positive expression rate of the two proteins was obviously higher in colorectal adenocarcinoma tissue(P<0.05). Expression rate of Caspase-3 was 48.14%(52/108), 65.00%(26/40), 95.00%(38/40), in the three kinds of tissues. The expression of Caspase-3 was lower in colorectal adenocarcinoma than in adenoma and adjacent mucosa(P<0.05).2. The high expression of protein EZH2 was related to neoplasm’s position, differentiateddegree, TNM stages and lymphatic metastasis(P<0.05), and was unrelated to age and sex.3. The expression of VEGF was related to TNM stages and lymphatic metastasis(P<0.05), and was unrelated to age, sex, neoplasm’s position and differentiated degree(P>0.05).4. The positive expression of Caspase-3 was related to neoplasm’s position, differentiated degree, TNM stages and lymphatic metastasis(P<0.05), and was unrelated to age and sex.5. In colorectal adenocarcinoma the expression of EZH2 and protein VEGF were positive correlation(r=0.257, P<0.05); the expression of EZH2 and protein Caspase-3 were negative correlation(r=-0.323, P<0.05); the expression of VEGF and Caspase-3 were negative correlation(r=-0.350, P<0.05)6. The result of Western Blot indicated that the expression of protein EZH2 in adenocarcinoma, adenoma and adjacent mucosa was 0.549±0.145, 0.283±0.023, and 0.107±0.022 respectively. Compared with adjacent mucosa tissue, the expression of EZH2 in adenocarcinoma and adenoma increased highly; the expression of EZH2 in adenocarcinoma was higher than in adenoma, and all of the differences above were statistically significant(P<0.05).Conclusion:1. The EZH2 gene was over expressed in colorectal adenocarcinoma tissues,and it was significantly associated with low differentiation, advanced clinicopathologic staging and metastasis.2. The correlation analysis showed that EZH2 and VEGF were positively correlated,both exhibited synergestic effect on colorectal cancer angiopoiesis;EZH2 and VEGF were negetively correlated,EZH2 gene may affect proliferation effects of cancer cell by inhibit apoptosis;EZH2 together with VEGF and Caspase-3 promote malignant progression of colorectal cancer.
