ObjectiveExplore the effects of aberrant expression of XIAP and Caspase-3proteinson the pathogenesis and progression in colorectal adenocarcinoma.MethodsXIAP and Caspase-3proteins were detected Using immuno-histochemistrymethod, and XIAP protein was detected by Western Blot technique incolorectal adenocarcinoma and adenoma.ResultsThe results of immuno-histochemistry:1、Positive rate XIAP proteinwere73.1%(49/67) in colorectal carcinomas, but30.0%(9/30) in adenomas;Lower positive rate of XIAP protein was observed in the group of well-moderate differentiation (66.7%) than that in the group of poorly-diffe-rentiation(93.8%), significant difference statistically was obtained between the twogroups;In the groups of Infiltration depth T1+T2period and T3+T4periodpositive rates of XIAP protein were70.0%and74.0%respectively,andpositive rates of XIAP protein were73.7%and65.5%respectively in the groupsof positive and negative lymph-node metastasis, but no significantdifferentiation statistically were found. Positive rate of caspase-3protein was 18.0%(12/67) in colorectal adenocarcinoma, but60.0%(18/30) in adenoma;In the group of well-moderate differentiation, higher positive rate of caspase-3(31.4%) was observed than that in the group of poorly differentiation (6.2%);Significant difference statistically was obtained between the two groups. In thegroups of Infiltration depth T1+T2period and T3+T4period, positive rate ofcaspase-3protein were12.5%(5/40) and11.1%(3/27) respectively, and5.3%(2/38) and10.3%(3/29) respectively in the groups of positive lymph nodemetastasis and negative lymph node metastasis, but no significantdifferentiation statistically were found.The results of the Western blot protein quantification: XIAP monoclonalantibody displayed the specific bands in57kD. Higher level expression of XIAPprotein was found in colorectal cancer than that in colorectal adenoma andmucosa adjacent to the cancer, modetately differentiated and poorlydifferentiated group with significant difference, no significant differentiationstatistically were observed between the groups of lymph node metastasis andwithout lymph node metastasis,the groups of Infiltration depth T1+T2periodand T3+T4period.ConclusionsOver-expression of XIAP protein and lower-expression of Caspase-3protein might be play an important role in pathogenesis of colorectal carcinoma,but no significantly difference statistically were obtained between it’s expressionand the malignant progression in colorectal adenocarcinoma.
