Objective: To investigate the expression and significance of apoptosis-related gene Livin,Caspase-3 and Bcl-2 in colorectal adenocarcinoma.Methods: Utilizing high-throughout tissue microarrays and immunohistochemical streptavidin-perosidase,the expression of Livin,Caspase-3 and Bcl-2 was detected in 115 cases of colorectal adenocarcinoma,19 cases of colorectal adenoma and 12 cases of normal colorectal mucosa.And we analyze the relationship between these factors and clinicopathologic characteristics according to the survival time of 71 cases which were follow-up from 6 months to 6 years.Results: The positive expression of Livin in normal mucosa was 8.33% lower than 10.53% in adenoma(p>0.05) and 49.57% in cancer specimens(P<0.05). The positive expression of Caspase-3 in normal mucosa was 83.33%, higher than 47.37% in adenom(ap<0.05) and 41.74% in cancer specimen(sP<0.05).The positive expression of Bcl-2 in normal mucosa was 25%,lower than 84.21% in adenoma(p<0.05) and 58.26% in cancer specimens(P<0.05). The expression of Livin,Caspase-3 and Bcl-2 was significantly correlated with degree of differentiation(p<0.05),and Livin's expression was also related to lymph node metastasis and tumour infiltrating degree(p<0.05). Spearman rank correlation showed a significant cooperativity among Livin, Caspase-3 and Bcl-2 protein (P<0.01). The positive expression of Livin and negative expression of Caspase-3 were significantly related to shot survival time(p<0.05). Cox multivariate analysis revealed that Livin and Caspase-3 were independent prognosticators for colorectal adenocarcinoma (p<0.05).Conclusion: Tissue microarray technique is an effective method to detect multiple gene protein expression. Livin,Caspase-3 and Bcl-2 may play an important role in genesis development and metastasis of colorectal adenocarcinoma.
