Objective To study the expressions and significance of Raf-1 protein kinase and VEGF in colorectal adenocarcinoma, colorectal adenoma and corresponding para-cancer normal colorectal tissues.Methods To collect 112 samples from 72 cases of colorectal adeno- carcinoma (including 72 cancerous tissues and 40 para-cancerous normal colorectal tissues), and 40 samples from 40 cases of colorectal adenoma. The expressions of Raf-1 protein kinase and VEGF were detected by immunohistochemical staining.Results The positive percentage of Raf-1 protein kinase was 29.17% in colorectal adenocarcinoma, 2.50% in corresponding para-cancer normal colorectal tissues and 12.50% in colorectal adenoma. The positive percentage of VEGF was 41.67% in colorectal adenocarcinoma, 10.00% in corresponding para-cancer normal colorectal tissues and 22.50% in colorectal adenoma. The positive percentage of Raf-1 protein kinase and VEGF was significantly higher in colorectal adenocarcinoma tissues than in colorectal adenoma tissues and corresponding para-cancer normal colorectal tissues (P < 0.05). The expression of Raf-1 protein kinase in the colorectal adenocarcinoma was not associated with age, sex, lesion size, histological classification, depth of invasion, lymph node metastasis and Dukes stage(P>0.05). The expression of VEGF was not associated with age, sex, lesion size, histologyical classification(P>0.05),but was associated with depth of invasion, lymph node metastasis and Dukes stage in the colorectal adenocarcinoma (P<0.05).Conclusion Raf-1 protein kinase and VEGF promote the development of the colorectal adenocarcinoma. It is not sure that Raf-1 protein kinase has much value in the estimation of the invasion and metastasis of colorectal adenocarcinoma. VEGF is close related to angiogenesis of colorectal adenocarcinoma; also, VEGF promotes colorectal adenocarcinoma's growth, invasion and metastasis.
