【Objective】To explore the expression and clinical effects of COX-2, VEGF and MMP-2 protein in colorectal adenocarcinoma, and detect the roles of these factors in the development of colorectal adenocarcinoma.【Methods】Immunohistochemical SP staining method was used to detect the expression of COX-2, VEGF and MMP-2 in 53 cases of colorectal adenocarcinoma, 12 cases of colorectal adenoma and 10 cases of normal colorectal mucosa. And we analyze the relationship between these factors and clinicopathologic characteristics, and the relationship of the factors.【Results】1. In clinical samples the positive rates of COX-2 in colorectal adenocarcinoma, colorectal adenoma and normal colorectal mucosa were 83.0%(44/53), 75%(10/12) and 20%(2/10), respectively. Compared with the positive rate of normal colorectal mucosa, the positive rates in every other group showed significant difference(P<0.01, P<0.01). The positive rates of VEGF in colorectal adenocarcinoma, colorectal adenoma and normal colorectal mucosa were 86.8%(46/53), 66.7%(8/12) and 30.0%(3/10), respectively. Its positive rate showed significant difference(P<0.01) between colorectal adenocarcinoma and normal colorectal mucosa. The positive rates of MMP-2 in colorectal adenocarcinoma, colorectal adenoma and normal colorectal mucosa were 71.7%(38/53), 66.7%(8/12) and 20.0%(2/10), respectively. Compared with the positive rate of normal colorectal mucosa, the positive rates in every other group showed significant difference(P<0.01, P<0.05). Spearman correlation analysis showed that there was a significant positive correlatin between the expression of COX-2 and VEGF, COX-2 and MMP-2, VEGF and MMP-2, respectively(P <0.05).2. The expression of VEGF in 53 cases of colorectal adenocarcinoma was significantly correlated with the degree of differentiation and the depth of infiltration(P<0.05, P<0.01). The expression of MMP-2 was significantly correlated with the degree of differentiation in 53 cases of colorectal adenocarcinoma(P<0.05). The expression of COX-2 in 53 cases of colorectal adenocarcinoma was not correlated with sex and age of sufferers, type of the tumour, size of the tumour, the degree of differention, the depth of infiltration, TNM staging and the lymph node metastasis.【Conclusions】The fact that COX-2,VEGF and MMP-2 over-express in colorectal adenocarcinoma suggests that they may be regarded as markers of the development and progression of colorectal adenocarcinoma. A significance positive correlation between COX-2 and VEGF suggests COX-2 may up-regulate the expression of VEGF to promote the development and progression of colorectal adenocarcinoma. The significant positive correlation between COX-2 and MMP-2 suggests COX-2 may induce the expression of MMP-2 to promote the development and progression of colorectal adenocarcinoma. The positive correlation between VEGF and MMP-2 shows a significant cooperativity between the two factors in the development and progression of colorectal adenocarcinoma. The expression of VEGF and MMP-2 are significantly related to some clinicopathologic characteristic in colorectal adenocarcinoma.
