Object: Our study aimed to comparative study of the two groups, which includes pre-embryo stops growing by patients and the normal pregnancy group, the following data were been collected, 1) the serum concentrations of IL-10 and TGF-β1, 2) TGF-β1 and IL-10 m RNA expression levels in villous tissue, 3) protein expression levels of TGF-β1 and IL-10 in villous tissues, through conducted the study, we proposed to clarify the differences of the TGF-β1 and IL-10 both in serum and in villous tissues between the embryo development stop patients group and the normal pregnancy pregnant women group.Methods: 1.ELISA assay were conducted to detect the TGF-β1 and IL-10 concentrations in serum;2.Immunohistochemical detection of villi TGF-β1 and IL-10 in the protein expression levels;3.RT-PCR amplification was used to detect the TGF-β1 and IL-10 m RNA expression levels.Results: 1.ELISA assay experimental group and control group in serum TGF-β1 IL 10-concentration and, The experimental group were higher than the control group, there was significant differences.2.Immunohistochemistry(SP) method, morphological image analysis system to measure human chorionic tissue TGF-β1 and IL-10-positive cells AOD values, the results showed that, the number of positive cells amounts and TGF-β1 and IL-10-positive cells in the control group were higher content than the experimental group.3.RT-PCR amplification was detected in the TGF-β1 m RNA and IL-10 m RNA expression levels between the experimental group and the control group in villi tissues, the results showed that: the control group, higher levels of TGF-β1 m RNA and IL-10 than the experimental group There were significant differences between the two groups.Conclusion: The lower expressions of the immune cytokine TGF-β1 and IL-10 during pregnancy, may lead to the occurrence of embryo stops growing; conversely, The higher expressions of the immune cytokine TGF-β1 and IL-10 during pregnancy, played an important role in maintaining a normal pregnancy.
