Objectives:Intermedin(IMD) is a newly discovered member of calcitonin gene-related peptide family, which have a wide range of physiological functions similar to adrenomedullin in the central nervous system, cardiovascular system. The purpose of this s tudy is to verify whether injected exogenous IMD can promote recovery of blood flow and angiogenesis on ischemic organization of the acute/chronic lower arterial ischemia model in mice(completely/partially blocked femoral arteries).Methods:Mice(C57BL/6) femoral artery is completely blocked/partial suture blocked by surgery of acute/chronic lower arterial ischemia model,and establish the disease control group, fake-operation control group and blank control group.We use doppler blood flow imaging to assess the blood flow of mice model.After the operation,we give exogenous IMD 150μg/kg body-weight for each mice immediately by muscle injection, every mice injected with the same exogenous dose IMD one time a day, lasts 2 weeks.Then we take ischemic lower limb skeletal muscle as tissue samples. Record ing doppler blood flow changes on pre-operative,post-operative,and every day after the operative for 2 weeks. Skeletal muscle specimens respectively perform HE pathological staining and CD31,CD105 immuno- histochemical staining, which can measure microvascular density of marked by CD31,CD105.Results:When the acute and chronic lower limb arterial ischemia model in mice esta blished, the signal of lower limb blood flow o n doppler blood flow imaging redu ced. After IMD injection and as time goes post-operation, ischemic limb blood fl ow signal substantially restored, and recovery of blood flow signal in IMD injecti on group is significantly higher than that compared with the control group. Pathol ogic HE staining(x100) of lower limb ischemia skeletal muscle showed that the capillaries, the number of single or grouped e ndothelial cells around a blood vess els in IMD group is more than that in the control group,within the ischemic orga nization. Results of C D31, CD105 immuno- histochemical stainings marked microve ssel density showed that the IMD injection group was statistically significant diffe rence(P<0.05) compared to the control group.Conclusions:In the mice model of acute and chronic lower limb arterial ischemia, exogenous IMD injected intramuscular can promote recovery of blood flow in ische mic tissue and increasing angiogenesis endothelial cells density marked by CD31/C D105 immuno-histochemical staining, conforming that exogenous IMD can promote recovery of blood flow and microvessel angiogenesis, which conducive to the neovascularization of the ischemic tissue.
