Aim:Observe whether Astragaloside IV joint remote ischemic postconditioning promote angiogenesis effect of lower limb ischemiaObjective:40 male SD rats were randomly divided into 5 groups:sham group (n=8)、model group (n=8)、AS-IV group (n=8)、RIPOST group (n=8)、combined group (AS-IV+RIPOST) (n=8);AS-IV were intragastric administrated for 2weeks (50mg/kg/d);rats in RIPOST group receive 3 cycles of 10 min left limb ligation followed by 10min reperfusion in one day for 2weeks;combined group receive both AS-IV and RIPOST for 2 weeks; equal normal saline were intragastric administrated every day in model group, sham group and RIPOST group.Rats were sacrificed 2 weeks after the intervention, whichever gastrocnemius muscle tissue, part of the pathological sections were HE stained section of CD31 expression in the ischemic tissue detected by immunofluorescence method, the measured density of new blood vessels. Western Blot method to detect p-AKT, protein expression of VEGF.Results:Compared with model group, AS-IV group, RIPOST group were improved severity of ischemic tissue, and promote local angiogenesis (p<0.05);Results combined intervention group than AS-IV group, RIPOST group improved more obvious (p<0.05).Tip RIPOST joint use with AS-IV can be further reduced after infarction gastrocnemius degeneration and necrosis, reduce fibroblast proliferation and increase in inflammatory cells.And AS-IV group, RIPOST group were able to increase vascular endothelial growth factor expression and promote regional ischemic angiogenesis and improve perfusion, joint intervention group stronger effect.Conclusion:AS-IV treatment, RIPOST can be adapted to improve the blood supply to the ischemic hindlimb function, reduced infarct size gastrocnemius, promote angiogenesis and improve regional ischemia reperfusion;AS-IV therapy can enhance the protective effect RIPOST lower limb ischemia in rats.
