Object: Intestinal tract has many functions that include not onlydigestion, absorption and peristaltic movement, but immunoloregulation, hormone secretion, mucous membrane barrier function as well. Normal barrier function that is an important characteristic of the gut depends on the effective working of epithelial, molecular, and immune components. Among them, the mucosa epithelia barrier plays the most important role. Under the conditions of trauma, operation, radiation, chemotherapy and severity infection, the secondary ischemia and hypoxia can result in ischemia-reperfusion injury. Then the barrier dysfunction caused by the detrimental factors including oxygen free bases and inflammation was formed.At present, the treatments of the barrier dysfunction comprehend amelioration hemoperfusion of the intestinal epithelium, application of selective intestinal tract detergent, nutritional support and Chinese medicine.The functions of fragrant drugs include promoting appetite, facilitating resorption and utilization of nutrient, rising exponent of thymus gland and spleen of mice, reinforcing activity of macrophage,plerosis cell membrane and cellular skeleton, inhibiting injury of intestinal mucosa by various agent. Fragrant drugs have been proven to protect intestinal mucosa with nonspecific role. It may be relating to adjust and protect the barrier structure and function directly. Fragrant drugs containing rich volatile oil that can eliminate wetness have such functions as dispersing vital energy, eliminating wetness, invigorating the spleen and the stomach. Being one of the main and commonly used fragrant drugs in Chinese Medicine, Huoxiang can activate the spleen therapy, eliminate the wetness-evil, dredge the middle warmer, recover the reception and transportation of the spleen and stomach. Guanghuoxiang (cablin potchouli herb) contains 1.5% of volatile oil whose main active component is patchonlialalcohol, about 52%~57% of the total amount. Clinical results indicated that Xiaogantang whose main component is Huoxiang significantly increase the intake of meals, ameliorate the level of nutrients in body; enhance immunity, and decrease infectious rate. But the state of the art research only environ machine barrier, so it hard to reveal mechanism of Chinese medicine integrally. The purpose of the study is to examine the effect of HuoXiang on protecting intestinal barrier function through lower limb ischemic-reperfusion model induced - from Wistar rats. The pathological changes of the intestinal structure were observed underelectron microscope. The serum NO,TNF- a level, DAO activities of plasma and membrane fluidity of intestinal epithelial cell (IEC) wereexamined. Methods:1 Lower limb ischemic-reperfusion model was induced from Wistar rats. Groups with different herb concentrations were created.2 Observing mucus under HE and ABPAS staining; mast cell and reticular fiber were studied under toluidine blue and Gomori staining respectively. Examining the DAO activities of plasma. The small intestinal histopathology and ultrastructural of rats were observed with optical microscope and electronic microscope respectively.3 Examining the serum NO,TNF- a level, DAO activities of plasmaand membrane fluidity of intestinal epithelial cell.4 Quantitatively analyzing the area of mucus and number of mast cell through image analysis system.5 Examining the membrane fluidity of intestinal epithelial cell. Results:1 Of the rats with IR, vascular congestion, intestinal edema, infiltration of inflammatory cells and exfoliation of epithelia cell in the ileal mucosa were produced. Massive aggregation of polymorphonuclear leukocytes within the lamina propria were observed in experimental group. Mitochondria swelling, rough endoplasmic reticulum expanding under TEM.2 The concentration of serum TNF- a and NO significantlyincreased in the model group, so did the activity of DAO. The membrane fluidity of intestinal epithelial cell decreased.
