Objective: To investigate the application of peripheral blood PCA3 gene expression in early diagnosis of prostate cancer.Methods: Quantiative Real-time PCR(Taqman probe) was applied to detect peripheral blood PCA3 gene expression of patients with prostate cancer,benign prostatic hyperplasia, chronic prostatitis and healthy adults. PCA3 expression level was compared between four groups. The relationship between PCA3 m RNA expression and Gleason scores was evaluated. The difference of PCA3 m RNA expression before and after endocrine therapy treatment was compared.Results: Peripheral blood PCA3 gene expression was only detected in prostate cancer group(31/46) and benign prostatic hyperplasia group(4/40)with no expression in other groups and the expression level was higher in non-localized prostate cancer group than that in localized prostate cancer group. There was no significant difference in PCA3 gene expression in patiens with prostate cancer of different Gleason scores. The level of PCA3 gene expression was lower after endocrine therapy treatment than that of before treatment.Conclusion: PCA3 gene was a highly specific molecular marker for prostatecancer and peripheral blood PCA3 gene expression had a promising clinical application in early diagnosis of prostate cancer.
