Objective:The purpose of this study was to investigate PCA3 gene expression in peripheral blood of prostate cancer(PCa)and benign prostate hyperplasia(BPH),and further to assess its clinical significance.Methods:We determined the copies of PCA3 mRNA in peripheral blood of PCa and BPH from131 samples(PCa,n=78;BPH,n=53)using a quantitative reverse transcriptase-quantitative polymerase chain reaction(RT-qPCR)with TaqMan assay.The difference between PCA3,PSA and fPSA in prostate cancer group and prostatic hyperplasia group was compared and analyzed firstly.The results of PCA3,PSA and fPSA in prostate cancer patients were further studied,and the ROC curves were plotted respectively.The area under the curve(AUC)was calculated and the PCA3 mRNA copies of peripheral blood and tPSA and fPSA on prostate cancer were compared and analyzed.The difference between the sensitivity and specificity of the diagnosis.The relationship between the expression of PCA3 gene in peripheral blood and the Gleason score and the TNM stage of the prostate tumor were analyzed.The clinical significance of the expression of the prostate cancer in the early diagnosis of prostate cancer was discussed.Results:(1)The clinical data revealed that expression of PCA3 gene was significantlyhigher in PCa than in BPH,The difference was statistically significant,P<0.05.The area of PCA3,PSA and fPSA under the curve(AUC)was 0.738,0.570 and 0.554,respectively.The area under the PCA3 curve was significantly larger than that of PSA and fPSA,P<0.05,it indicates that the expression level of PCA3 mRNA can predict prostate cancer better.(2)The expression of PCA3 mRNA in peripheral blood of prostate cancer patients with Gleason score of more than 8 was significantly higher than that of patients with Gleason score less than 7.(3)The expression of PCA3 gene in peripheral blood after radical resection of colorectal adenocarcinoma was significantly lower than that before operation.Conclusion: Increased PCA3 in peripheral blood is correlated with PCa,and the detection of PCA3 may significantly reduce adverse screening outcomes.PCA3 gene expression in peripheral blood had a promising clinical application in the early diagnosis of PCa.
