Objective: Research on the value about peripheral blood prostate cancer antigen 3(PCA3)in prostate specfic antigen(PSA)grey area(4-10 ng/ml)to the diagnosis of prostate cancer.By measuring PCA3 expressed in prostate cancer patients with different pathological grading to study the relationship between the PCA3 and the pathologic grading of prostate cancer(differentiation).Methods: Randomly selected 30 patients with prostate cancer from the urology department,first hospital of Shanxi Medical University from April to December 2016 as experimental group.Another selection of 34 patients with BPH as control group,and their PSA for 4-10ng/ml.Using RT-PCR technique to detect the relative expression of PCA3 m RNA in two groups of blood mononuclear cells,and statistical the relationship between the well differentiated(Gleason score 2-4),medium differentiated(Gleason score 5-6)and poorly differentiated(Gleason score 7-10)prostate cancer in prostate cancer group and PCA3 m RNA relative expression quantity.Results: In 64 cases of subjects,30 cases of pathological diagnosed with prostate cancer,the relative expression of PCA3 was 2.50 ± 1.79;34 cases of benign prostatic hyperplasia,the relative expression of PCA3 was 1.01 ± 0.54.P < 0.05,the difference was statistical significant.In 30 cases of prostate cancer patients,8 cases of Gleason score are 7-10 points,the relative expression of PCA3 was 4.40 ± 2.27;17 cases of Gleason score are 5-6 points,the relative expression of PCA3 was 2.13 ± 0.76;5 cases of Gleason score are 2-4 points,the relative expression of PCA3 was 0.72 ± 0.28.Pair-wise comparison difference between three groups have statistical significance,show that the relative expression of PCA3 in poorly differentiated prostate cancer patients is higher than that of well differentiated and medium differentiated ones,the relative expression of PCA3 in medium differentiated prostate cancer patients is higher than that of well differentiated.Conclusion: In patients with PSA in the grey area,the higher the PCA3 m RNA relative expression,the greater the risk of prostate cancer which can provide reference for prostate cancer screening,prompting that biopsy positive rate is high.In prostate cancer patients with PSA in the grey area,the higher the relatively expression of PCA3 m RNA,the higher the prostate cancer pathology grading,the greater the prostate cancer cells malignant degree,the lower of differentiation,prompting with poor prognosis.
