Objective:The current most commonly used tumor marker indiagnosis of prostate cancer is the prostate specific antigen(PSA),however, there is still controversy over its value in early diagnosis ofprostate cancer. The recently found non-coding prostate cancer gene3(PCA3)is the most specific biomarker in diagnosing prostate cancer(PCa).Whereas the clinical value of PCA3is proved in diagnose usingprostate massage urine, there are still very few researches on its clinicalvalue in PCa diagnosis using peripheral blood lymphocyte(sPBL). In thisstudy, a method of detecting expression of PCA3in blood serum isproposed as an alternative to diagnosis of prostate cancer.Methods:From June2012to November2013, specimen of peripheralblood and prostate tissue were collected from hospitalized patients in bighospitals located in north and middle part of Guangxi, of which28casesare tissue and peripheral specimen from confirmed PCa patients,34casesare specimen from benign prostatic hyperplasia(BPH), and31cases ofspecimen from normal people without the disease were used as normalcontrol.The expression of mRNA of PCA3gene in patents’ peripheral blood and prostate tissue were investigated via RT-PCR first, and thendifference in expression of RNA of PCA3among different groups(prostate cancer group, BPH group and normal control group) wereanalyzed.The relationships among mRNA expression of PCA3in PCa,Gleason scoring and tumor TNM stage were investigated, and the effectof both the expression and the PSA level on prostate cancer diagnosis isanalyzed.Results:After detection and analysis, it is found out that the expressionof PCA3in above-mentioned3groups is statistically differen(tP<0.005),with expression of PCA3in tissue and peripheral blood of PCa groupsignificantly higher that of the rest2groups. There is no significantdifference in expression of PCA3in BPH group and the control group(P>0.05)。Using expression of PCA3in blood for prostate cancerdiagnosis, the specificity and sensitivity of which are96.9%and32.1%,respectively, if combined with test of PCA3in prostate tissue, will beimproved. At the same time, detection of expression of PCA3in bloodcoupling with PSA≥10ng/ml offers95.4%in specificity and60.7%insensitivity.Conclusion:1. The expression of mRNA of PCA3gene in peripheralblood and tissue is a specific indicator for prostate cancer diagnosis,mRNA of PCA3in patient’s blood can be considered as tumor maker inscreening for PCa.2. Detecting mRNA expression of PCA3in peripheralblood together with PSA≥10ng/mL can, not only, improve the sensitivityand specificity in prostate cancer diagnosis, but also reduce unnecessary invasive examination and prevent misdiagnosis.
