Objectives This study adpot the case-control method. In Chinese han population, to explore the relationship between polymorphism of IL-17(three gene loci)and the susceptibility of silicosis through analyzed the genotype of IL-17A(rs2275913, rs4711998) and IL-17F(rs763780)in pneumoconiosis case group and control group. In addition, combined the linkage disequilibrium tests and haplotype analysis between IL- 17 gene loci with the analysis results of three gene loci of IL-17, to explore the relationship between haplotype and the susceptibility of silicosis. At last, a preliminary study is used to explore the interaction between gene loci.Methods Establish the database by Excel 2003 and using SPSS 17.0 statistical software to analyzed the data processing and Statistical analysis. The Polymorphisms of IL-17 in219 cases and 242 controls were evaluated by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP). Applied online SHEsis software for each locus linkage disequilibrium analysis or haplotype analysis. Multifactor Dimensionality Reduction(MDR) Used for the analysis of interactions between genetic loci.Results The allele frequencies distribution of three gene loci at rs2275913,rs4711998 of IL-17 A and rs763780 of IL-17 F sites in controls were conformed to Hardy-Weinberg equilibrium(P>0.5), this result shows that the selected samples have good group represen-tative. There were no differences of age, smoking, the dust accumulated by length of service and type of work between pneumoconiosis case group and control group(P>0.5).rs2275913 of IL-17 A has enzymed into AA, AG and GG genotype, their gene frequencies in pneumoconiosis case group were 30.1%, 53.9% and 16.0%, and the gene frequencies in control group was 31.0%, 47.9% and 21.1%, with no significant difference between three kinds of genotypes in the two groups(P>0.05). rs4711998 of IL-17 A has enzymed into AA, AG and GG genotype,their gene frequencies in pneumoconiosis case group were 46.1%,47.0% and 6.9%,and the gene frequencies in control group were 47.9%,45.1% and 7.0%. there was no significant difference between pneumoconiosis case group and control group(P>0.05). The genotype frequencies of AA,AG and GG of IL-17 F rs4711998 in case group and control group were79.0%, 20.5%, 0.5%and67.8%, 38.1%,0.4%respectively, after statistical tests, there was a significant difference between three kinds of genotypes in the two groups(P<0.05).The allele frequencies of A and G in case group and control group were 89.3%, 10.7%and 83.7%, 16.3%respectively, the difference has statistical significance(P<0.05). After test to three gene loci at rs2275913,rs4711998 of IL-17 A and rs763780 of IL-17 F, there was no linkage disequilibrium between each other gene loci(D’<0.7,r2<0.3), haplotype analysis was tested to three genetic loci and built eight kinds of haplotypes, the frequency distribution of GAG haplotype has a significant difference between pneumoconiosis case group and control group, and the other haplotype has no significant correlation with the susceptibility of pneumoconiosis(P>0.5). All of three Gene loci in IL-17 A and IL-17 F were analyzed by the MDR method,The results showed that three genetic loci have no interaction between each other(P>0.5).Conclusions 1 No relationship was found between gene loci at rs2275913 or rs4711998 of IL-17 A gene polymorphisms and pneumoconiosis susceptibility in this study. 2 The gene loci at rs763780 of IL-17 F gene polymorphisms was associated with pneumoconios is susceptibility, AG genotype is the protection genotype of pneumoconiosis, and G allele is the protection allele of pneumoconiosis. 3 There was no linkage disequilibrium between three genetic loci in this study, GAG haplotype is the protection factor of pneumoconiosis. G allele of rs2275913 and A allele of rs4711998 could increasing the protective efficacy of pneumoconiosis of G allele at rs763780. 4 There is no interaction between rs2275913, rs4711998 and rs763780 gene loci in our study.
