Objective In order to explore the association between polymorphisms of epidermalgrowth factor (EGF) gene (G-61A, D784V, R431K) and the susceptibility ofpneumoconiosis, by case-control study. We try to provide evidence of genetic susceptiblemarkers of EGF gene for screening the high risk group of workers exposed to dust.Methods This study sample included2568coal workers and2265steel enterpriseworkers. The cases was228pneumoconiosis, including116silicosis and112CWP. Thecontrol was279non-pneumoconiosis.1.5ml venous blood was drawn by anticoagulants,stored in-20℃freezer. DNA was extracted from whole blood through the saltingmethod. The polymorphisms of EGF (three locus) were detected by the method of CR-RFLP techniques. Genotypes variants use the χ2test.Results1In general The pneumoconiosis average age was55.3±8.0years, averagecumulative dust exposure was29.4±6.5years. The control was55.0±7.1years and30.1±5.4years. The silicosis average age was58.9±7.9years, average cumulative dustexposure was28.9±6.3years. The control was57.2±7.5years and29.7±5.8years. TheCWP average age was51.5±6.4years, average cumulative dust exposure was29.8±6.7years. The control was52.4±1.9years and30.6±4.7years. All factors were notsignificance (P>0.05).2The frequencies of G-61A locus GG, GA and AA in case were51.3%,35.5%and13.2%, compared with the control (36.6%,45.2%and18.2%), thedifference was significant(P<0.05). The frequencies of allele were69.1%,59.1%for Gin two groups, allele were30.9%,40.9%for A in two groups. No significant associationsbetween groups(P>0.05). The frequencies in the silicosis were50.8%,34.5%and14.7%,compared with the control (35.6%,44.3%,20.1%), the difference was significant(P<0.05). The frequencies of allele G in two groups were68.1%,57.7%, allele A in silicosisand control were31.9%,42.3%, there was significant difference (P<0.05). The genotypefrequencies in the CWP were51.8%,36.6%and11.6%, compared with the control(37.6%,46.2%and16.2%), the difference was no significant(P>0.05). The frequenciesof allele G in case and control were70.1%and60.8%, allele A in CWP case and controlwere29.9%,39.2%, there was significant difference (P>0.05).3The frequencies ofD784V locus AA, AT and TT in case and control were56.6%,36.8%,6.6%and65.6%,30.8%,3.6%, there was no significant difference (P>0.05). The frequencies of allele were 75.0%,81.0%for A in cases and controls, allele were25.0%,19.0%for T in case andcontrol. No significant associations between groups(P>0.05). The frequencies insilicosis and control were58.6%,34.5%,6.9%and65.1%,31.5%,3.4%, there was nosignificant difference (P>0.05). The frequencies of allele A in silicosis and control were75.9%,80.9%, allele T in case and control were24.1%,19.1%, there was no significantdifference (P>0.05). The frequencies in CWP and control were54.4%,39.3%,6.3%and66.2%,30.0%,3.8%, there was no significant difference (P>0.05). The frequencies ofallele A in CWP and control were74.1%and81.2%, allele T in case and control were25.9%,18.8%, there was no significant difference (P>0.05).4The frequencies of EGFR431K locus GG, GA and AA in case and control were55.7%,40.4%,3.9%and58.4%,37.3%,4.3%, there were no significant difference (P>0.05). The frequencies of allelewere75.9%,77.1%for G in case and control, allele were24.1%,22.9%for A in case andcontrol. No significant associations were found between groups(P>0.05). Thefrequencies in silicosis and control were56.9%,39.7%,3.4%and55.0%,39.6%,5.4%,respectively, there were no significant difference (P>0.05). The frequencies of allele were76.7%,74.8%for G in silicosis and control, allele were23.3%,25.2%for A in silicosisand control. No significant associations between groups(P>0.05). The frequencies inCWP and control were54.4%,41.1%,4.5%and62.3%,34.6%,3.1%, there were nosignificant difference (P>0.05). The frequencies of allele were75.0%,79.6%for G inCWP and control, allele were25.0%,20.4%for A in CWP and control. No significantassociations between groups(P>0.05).Conclusions1The genetic polymorphism of G-61A relatied to susceptibility ofpneumoconiosis of carrying GG genotype.2No relationships between D784V genotypesand pneumoconiosis had been found in this study. But relationships between allele andpneumoconiosis had been found.3The relationship between genetic polymorphism ofEGF R431K and pneumoconiosis hasn’t been found in the study.
