Objective:Ophthalmic pterygium is one of common ocular surface disease, Palpebral area showed hypertrophy of the bulbar conjunctiva and lower triangular fibrovascular tissue thickening, its head invade cornea, its body on one side of palpebral area. When pterygium tissue hypertrophy, and Vasodilation growth to the central cornea, could cause corneal astigmatism, even lead to decreased of vision and affect the normal eye movement. Studies show that pterygium is one of conjunctival tissue disease, its pathogenesis is still unclear, may be related about abnormal cell proliferation and apoptosis.Pterygium is derived from the limbal epithelial stem cells abnormal proliferation and inflammatory growth, Fibrovascular tissue invasion corneal layer, Resulted in a wide range of matrix remodeling. Ki-67 antigen is an antigen expressed in proliferating cells, distributed in the nucleus. And its features is considered related about connected with chromatin and the mitotic closely. Some people think it may be having an important structural protein binding properties. In mitosis function, maintain important role in the structure of DNA rules. Is a sensitive indicator of cell proliferation reflected.PCNA is a nuclear protein required for eukaryotic cell DNA synthesis, and Its volume changes consistent with the DNA synthesis. Therefore PCNA is considered closely related to proliferation activities. detecting its expression in cell, Can be used as an accurate, simple and intuitive research evaluating an important indicator of the state of cell proliferation. In this study, immunohistochemical detection of proliferating cell nuclear antigen Ki-67、Cytokeratin CK3, proliferating cell nuclear antigen PCNA expression in normal conjunctiva and pterygium. Through their respective expression, to investigate thecharacteristics of the three clinical cases with pterygium relationship and their respective correlations between analyzed and summarized to further explore the pathogenesis of pterygium. Methods:Immunohistochemical SP method detected 30 cases of normal conjunctival tissue,45 cases of pterygium of proliferating cell nuclear antigen Ki-67, cytokeratin CK3,proliferating cell nuclear antigen expression of PCNA explore Ki-67, CK-3, PCNA expression in normal conjunctiva and pterygium tissue, and the correlation between their respective summarized.Results:1.Ki-67 there are 37 cases of pterygium tissue expression, the positive rate was82.2%, significantly higher than normal conjunctival tissue(9 cases, 30.0%), the difference was statistically significant(P <0.05).2.CK3 has 34 cases of pterygium tissue expression, the positive rate was 75.6%,significantly higher than normal conjunctival tissue(10 cases, 33.3%), the difference was statistically significant(P <0.05).3.There PCNA in 38 cases of pterygium tissue expression, the positive rate was84.44%, significantly higher than normal conjunctival tissue(6 cases, 20.0%), the difference was statistically significant(P <0.05).4.By analysis of the Spearman correlation found that:CK3 and Ki-67 expression in the conjunctiva and pterygium tissue showed a positive correlation, correlation coefficient r2 = 0.534, P = 0.038 <0.05; CK3 and PCNA expression in conjunctiva and pterygium tissue showed a positive correlation, r2= 0.214,P = 0.041 <0.05; Ki-67 and PCNA expression in the conjunctiva and pterygium tissue showed a positive correlation, r2 = 0.367, P = 0.007 <0.05.Conclusion:1.The experimental results show that Ki-67 expression in pterygium was significantly higher than normal conjunctival tissue, prompting the presence of abnormally high pterygium tissue proliferation, is a tissue disease.2.CK3 expression in pterygium was significantly higher than normal conjunctivaltissue, prompting the presence of abnormally high pterygium tissue proliferation, is a tissue disease.3.The experimental results show that the expression of PCNA in pterygium was significantly higher than normal conjunctival tissue, further proof of the existence of cells and the development of abnormal proliferation of pterygium, pterygium tissue have similar biological characteristics of the tumor.4.Ki-67 and CK3, PCNA expression in pterygium display, three were positive expression was significantly higher than normal conjunctival tissue was significantly positively correlated, Ki-67 with increasing CK3, PCNA expression and intensity increased, prompting three possible different ways to participate in the occurrence and development of pterygium tissue.
