[Objective] To investigate the effect of decorin on proliferation and apoptosis of human pterygium fibroblasts(HPF)in culture and compare the effect of MMC on human perygium fibroblasts,search for a new method to prevent the recurrence after pterygium surgery.[Methods] (1)Human pterygium fiborblasts were isolated from pterygium tissue caudomedial part of pterygium patients and cultured in vitro using tissue inoculation methods.(2)HPF cultured in vitro was treated with 0.01,0.1,1,5,10 mg/L DCN and MMC independly,afte 24h,48h,72h,observe HPF morphological alteration.The MTT method was used to assay the biologic activities of the two drugs in different doses after 12h,24h,48h, and compare the two drugs action.The expression of proliferating cell nuclear antigen(PCNA)in each group treated with DCN in different doses after 48h was detected by immuno-histochemistry method,the cell cycle distribution was measured by flow cytometic analysis.[Result] After administration of 10 mg/L DCN or 1 mg/L MMC for 12h,HPF could significantly be inhibited proliferation by the two drugs in a dose-and time-dependent manner(P<0.05). After treated with DCN 1-10mg/L for 48h,The number percentage of HPF in G0/G1 phase increased,while in S phase and increase rat(G2/M%+S%)decreased after treated with DCN 5-10mg/L for 48h (P<0.05),cells in late apoptosis were not found in control and groups.DCN (1-10 mg/L)can inhibit the expression of PCNA in HPF in a dose-dependent manner(P<0.05).[Conclusion] Decorin can significantly inhibit HPF proliferation,flow cytometic analysis show DCN block cells in G1 phase.
