Experimental Study On The Effect Of Huoxuesan In PGF2 And IL-6 In Rats With Acute Soft Tissue Injury

[Objective]:By establishing the rat model of acute soft tissue injury, we used Huoxuesan (the scattered ointment made by Nanjing Municipal Hospital of Traditional Chinese Medicine)as the external use drugs, measured the injury symptom index in rats before and after treatment, tested the local prostaglandin E2 (PGE2), interleukin 6 (IL 6) and morphology of local muscle tissue, preliminarily studied therapeutic action of Huoxuesan and its possible mechanism, to clarify the mechanism of analgesic effect, detumescence, and accelerating the speed of tissue repairment etc, to provide a theoretical basis for the clinical application of traditional Chinese Medicine.[Method]:The model rats were divided into 4 groups, each group had 16 rats:the normal control group (non model), model group (model, no treatment), Voltaren cream group, Huoxuesan group. No treatment was taken stright after the molding, corresponding drugs was given on the soft tissue lesions after 1 hour, observe limb symptoms everyday,and record the damage symptoms index. On the 3rd and 7th day, take 8 rats from each group, take the Orbital blood, separate the serum. Using ELISA method to determinate the prostaglandin E2 (PGE2), and the interleukin 6 (IL-6). After the blood-taking, the rats were sacrificed by cervical dislocation, took the local tissue for morphology observation.[Result]:3 and 7 days after the treatment, the damage symptoms index score of the Voltaren cream group and Huoxuesan group were significantly lower than the model group, had significant difference(P<0.01).The content of PGE2、IL-6 of the model group were significantly higher, compare with the normal control group(P<0.01). The content of PGE2、IL-6 of the Voltaren cream group and Huoxuesan group were significantly lower than the model group(P<0.01). After 7 days of treatment, the inflammation of the local soft tissue of the Voltaren cream group and Huoxuesan group were significantly reduced, the repairment of muscle fiber also relatively quicker than the model control group.[Conclusion]:Huoxuesan shows a good repairing effect on acute soft tissue injury of rat, can reduce the treatment, has functions such as relieving swelling and pain, anti-inflammatory, promoting the repairment of muscle fiber. The mechanism may be related to the inhibition of PGE2, IL-6 content of local injured tissues.
