ObjectiveBased on animal models of acute soft tissue injury of rabbits, with acutesoft tissue injury symptoms integral, leucocytes, platelet and histopathology asthe index. this thesis employs experiment method of modern pharmacology,and observes therapeutic action of acute soft tissue injury treated withtraditional Chinese medicine “Diechuanggao”.And then, after studyingpreliminarily this medicine from the point of PGE2and TNF-α, this paper aimsto provide experiment basis for clinical rational using medicine and lay thefoundation for developing new drugs.MethodsThere were486-month New Zealand rabbits (24male and24female),with weight2.5-3.0kg. After one week of adaptive breeding, they wererandomly divided into blank control group, model control group, traditionalChinese medicine safflower oil control group and “Diechuanggao”control group.Each group, consisting of12rabbits, had observation group of3days and7days.The right thigh furs of Rabbits’ were shaved24hours before theexperiment, with a scope of4cm×4cm. All the groups modeled according tothe related literatures except blank control group. Methods: after fixing theback body of rabbits, impact hammers fell down to the legs of rabbits from40cm15times, and each rabbit’s leg should be specified. Subcutaneous blood clots found with naked eyes after striking and swelling can be felt by handsindicated that models are established successfully. After modeling30minutes,rabbits were provided with medicines. As to “Diechuanggao” group,“Diechuanggao” is offered twice a day, with area of4cm×4cm. For safflower oilgroup, safflower oil was applied evenly on legs twice a day, with area of4cm×4cm, and then the wound should be wrapped with sterile gauzes. Withregard to the rest two groups, only sterile gauzes were offered. Local swelling,leukocyte, local tissue injury, PGE2content, serum TNF-αand other indicatorswere observed on the3rd and the7th day.Results1, The effect of “Diechuanggao” to the acute soft tissue injury of rabbitsAfter having medicines for3and7days, compared to blank control group,injury parts with obvious swollen, subcutaneous hemorrhage in many placesand injury parts with color of mulberry(P<0.01) in model control group. As for“Diechuanggao” group, the above indicators were better than blank controlgroup and got scores lower than model control group. From the aboveobservation, we know that “Diechuanggao” has the pharmacologic effect ofimproving acute soft tissue injury.2, The effect of “Diechuanggao” to leukocyte and Platelets of acute softtissue injury After having medicines for3and7days, the number of white bloodcell and Platelets in model control group are more than blank control group. Asto “Diechuanggao” group, the both number are lower than model control group,but better than safflower oil control group. These showed that “Diechuanggao”has the pharmacologic effect of reducing the number of leukocyte andPlatelets of acute soft tissue injury.3, The effect of “Diechuanggao” to pathology of acute soft tissue injury ofrabbits After having medicines for3and7days, there were some symptoms inmodel control group, such as subcutaneous tissue with blood, edema,inflammatory cell infiltrating, parts of muscular fibers broking, red-stained and band disappearing. There was notable hemorrhage in intercellular substancesand angiotelectasis with bleeding. Furthermore, the result of “Diechuanggao”control group is obvious better than model control group and safflower oilcontrol group. There was no significant change in blank control group. Afterabove analysis, we know that “Diechuanggao” has better recovery effect topathology of part acute soft tissue injury. See picture1, picture2, picture3,picture4, picture5, picture6, picture7, and picture8in the context.4, The effect of “Diechuanggao” to PGE2content as to acute soft tissueinjury of rabbits After having medicines for3and7days, the content of PGE2inmodel group was notable more than blank control group. The content of PGE2“Diechuanggao” control group, however, was obviously lower than modelcontrol group and better than safflower oil control group. So we can concludethat “Diechuanggao” could cure acute soft tissue injury through reducing thePGE2content.5, The effect of “Diechuanggao” to serum TNF-αof acute soft tissue injuryof rabbits After having medicines for3and7days, the content of TNF-αinmodel group was notable more than blank control group. But the content ofTNF-αin “Diechuanggao” control group is obviously lower than model controlgroup, and better than safflower oil control group. So we can say that“Diechuanggao” could cure acute soft tissue injury through reducing the serumTNF-α.Conclusions1, Traditional Chinese medicine “Diechuanggao” has better therapeuticaction for acute soft tissue injury. Specifically, it can improve injury symptom,reduce the number of leukocyte and Platelets, and promote pathological injury.2, The therapeutic action of traditional Chinese medicine“Diechuanggao”to acute soft tissue injury may through reduce PGE2andserum TNF-αcontent. 3, The therapeutic action of traditional Chinese medicine“Diechuanggao”to acute soft tissue injury is better than safflower oil.
