Objectives:To study the intervention effect of Kangshuan No.1 on lipid levels,vascular remodeling and endothelial injury factors in rats with atherosclerosis.Methods:Forty-two male SD rats were randomly divided into normal group(group a), model group(group b), positive drug group(group c), low dose group of chinese medicine(group d), middle dose group of chinese medicine(group e), high dose group of chinese medicine(group f)and combination of traditional chinese and western medicine group(group g),6 rats in every group. High lipid-diet and intraperitoneal injection of Vitamin were given to establish the atherosclerpsis(AS) model.Group c, group d, group e, group f, group g were intragastrically administered medicine from the nineth week. After 3 weeks,all the rats were sacrificed for deteminnation the levels of blood lipid,glutamic-pyruvic transaminase (GPT),glutamic-oxalacetic transaminase(GOT),low-density lipoprotein (ox-LDL),platelet endothelial cell adhesion molecule-1 (PECAM-1), platelet-derived growth factor(PDGF).The expression of ox-LDL,PECAM-1,PDGF were assayed by immunohistochemistry.The histomorphological changes of the aorta and livers were observed under light microscope.Results:(1) Compared with group a, levels of cholesterol(TC), LDL cholesterol(LDL-C), GPT GOT, ox-LDL,PECAM-1,PDGF were higher in group b (P<0.01); With forming a typical manifestation of atherosclerosis in aorta, the livers of rats in group b formed a manifestation of lipid deposition; (2) compared with the group b, the content of LDL-C、GPT、GOT、OX-LDL、PDGF、PECAM-1 in group d,e,f,g were lower (P<0.01, P<0.05),and levels of OX-LDL, PDGF were decreased with increasing the dose of Kangshuan NO.l;The foam cells were fewer,but the livers had no significant difference;The expression of OX-LDL), PDGF and PECAM-1 were lower (P<0.01);(3) Compared with group c,the content of PECAM-1 were lower in group g(P<0.01).Conclusion:(1) Kangshuan NO.1 could reduce the level of LDL-c and inhibit pathological vascular remodeling in rats with protecting the function of liver. (2) Reducing the expression of OX-LDL, PDGF, PECAM-1 may be the major mechanisms of anti-atherosclerosis of Kangshuan NO.1.
