Background and PurposeHepatocellular carcinoma(HCC) is a common malignant tumor in our country, there are many treatment methods such as liver resection, vascular intervention, radiotherapy, local ablation, we can perform one or combination therapy. Percutaneous chemical ablation is widely used because of it is a easy procedure, low cost, few side effects. Ethanol as a chemical ablation has been proved its effectiveness, but there are some shortcomings affect its clinical value for example repeated treatments, a range of side effects and so on.As a new type of sclerosant, Polidocanol (aethoxysklerol) has been widely used in European and America countries, and its domestic similar drug is lauromacrogol. At present, it is mainly used in varicose veins, vascular malformations, hemorrhoids, uterine fibroids, hydrocele, pulmonary bulla, cystic disease and so on, Studies have reported it can not only led normol liver cell degeneration and necrosis, but also inhibit the proliferation of human HepG2cells in vitro. Howerver there are no reports about its function on hepatocellular carcinoma in vivo as ethanol. So we set up the rabbit VX2liver tumor models to study the efficacy and safety about lauromacrogol.Materials and MethodsWith thirty-four rabbits were implanted with VX2tumors under laparotomy. Conventional CT scanning and enhanced CT scanning were used to check whether it successed or not after two weeks. After inoculation the experimental rabbits were randomly divided into three gourps of A(lauromacrogol injection), B(absolute alcohol injection), C(normal saline injection), and then performed the CT guided percutaneous chemical ablation. Serum alanine aminotransferase(AST), aspartate aminotransferase(AST), and blood urea nitrogen(BUN), creatinine(Cr) were detected 1day before treatment and1,3and7days after treatment, the general condition and body weight were also recorded. All rabbits were taken conventional CT scanning and enhanced CT scanning again14days after treatment before they were sacrificed.Then tumor volume,tumor growth rate were calculate and pathological changes were observed about the three groups of rabbits.ResultsThirty rabbit VX2liver tumor models were succseefully established verified by conventional CT scanning and enhanced CT scanning, The success rate was88%.Two rabbits died in the process of establishing of the VX2tumor model because of individual differences of the experimental rabbit to anesthesia, two rabbits did not find any VX2tumor in liver.Thirty rabbits were randomly divided into3groups, body weight and tumor volume before treatment in group A, B, C were(2.35±0.13) kg,(2.35±0.12) kg,(2.32±0.13) kg,(814±588.6) mm3,(904.5±549.4) mm3,(876.7±533.4) mm3, There were no statistically differences among the three groups in body weight and tumor volume (P>0.05).CT guided percutaneous chemical ablation were successfully performed in all of the experiments rabbits. There were no statistically differences among the three groups in ALT, AST, BUN, Cr before treatment. ALT levels were higher in each group at1day,3days,7days after treatment(P<0.05), AST levels were higher in each group at1day,3days after treatment(P<0.05), returned to pretherapy level at7days after treatment(P>0.05). No obvious change was observed about the level of BUN and Cr among the three gourps(P>0.05). Postoperative body weight and tumor volume in group A, B, C were (2.26±0.13) kg,(2.13±0.11) kg,(2.35±0.2) kg,(2306.4±1286.6) mm3,(2812.8±1560) mm3,(9122.9±9189.6) mm3. The body weight of group A, C had significant difference with group B(P<0.05), and the tumor growth rate of goup A, B had significant difference with group C(P<0.05).ConclusionPercutaneous puncture with lauromacrogol is not only a effective and safe method in treating VX2hepatic tumors, but also with limited side effects.
