| Objective: In this study,the Fibrosis Pigeon breeders’ lung of SD rats was made by using the exculpated protein such as pigeon fur.Under the intervention of Notch signaling pathway inhibitor DAPT,the protein expression differences of Notch signaling pathway ligand and receptor,as well as the mRNA expression differences of M1 type macrophage polarization marker gene(iNOS)and M2 type macrophage polarization marker gene(Arg-1)were detected.Hope can find in the Fibrosis Pigeon breeders’ lung of Notch signaling pathway and the relationship between macrophage polarization,deciphering the feeding pigeons in the lung macrophage polarization process influenced by what,may for the development of new therapies,looking for targeted therapy site intervention macrophage polarization direction,for he Fibrosis Pigeon breeders’ lung provides theoretical basis for accurate treatment.Methods : The 36 SD rats that passed the test were completely randomized and divided into ABCD 4groups by using random number table,among which,9 rats in group A and B were treated with sensitizer stimulation,9 rats in group C and D were treated with normal saline stimulation.The rats were cultured together for 20 weeks.Group B and C were again intraperitoneally injected with Notch signaling pathway inhibitor DAPT for in vivo experiment.CT and pathological section examination were performed to confirm fibrosis changes,lung tissues and cells were extracted immediately,and the protein expression of Notch signaling pathway ligand and receptor was tested by Western Blot.The mRNA expression levels of iNOS and Arg-1 in M1 type macrophages were detected by fluorescence quantitative PCR,and the differences in mRNA expression levels between each group were compared.Meanwhile,the effects of Notch signaling pathway blocking on Notch signaling pathway and the polarization direction of macrophages were analyzed.Results: At 20 weeks,2 rats in group A were randomly selected for CT and pathological examination,and typical pulmonary fibrosis changes were found in all rats,suggesting successful modeling.Meanwhile,the mRNA relative expression of iNOS gene was significantly different among groups A,B,C and D(F=2993.721,P=0.000).After multiple comparisons using LSD method,the relative expression level of iNOS mRNA in group A was higher than that in group B,C and D,and the difference was statistically significant(P<0.05).The relative expression level of iNOS mRNA in group B was higher than that in group C and D,and the difference was statistically significant(P<0.05).There was no significant difference in the relative expression level of iNOS mRNA between group C and group D(P>0.05).At the same time,there were statistically significant differences in the mRNA relative expression levels of Arg-1 gene among groups A,B,C and D(F=64.039,P=0.000).After multiple comparisons using LSD method,it was found that there was no statistical significance in the relative expression level of Arg-1 mRNA between group A and group B(P>0.05).The mRNA relative expression level of Arg-1 in group A and B was higher than that in group C and D,and the difference was statistically significant(P<0.05).There was no significant difference in the mRNA relative expression of Arg-1 between group C and group D(P>0.05).In this study,in detecting the protein expression content of each ligand and receptor in Notch signaling pathway,group A was the highest,group C was the lowest,and group B was between group D and group A,the control group.Further analysis of variance showed that the protein contents of Notch-related ligands and receptors were significantly different among groups A,B,C and D after different interventions(P<0.01).Conclusion: M1 type and M2 type polarization were found in macrophages of the Fibrosis Pigeon breeders’ lung of SD rats and normal rats,and the polarization of macrophages towards M1 type and M2 type macrophages increased in the Fibrosis Pigeon breeders’ lung of SD rats.Notch signaling pathway may be involved in the process of macrophage polarization to M1 type in the Fibrosis Pigeon breeders’ lung of SD rats.Macrophage polarization to M2 type was not interfered.In this study,we further understand the polarization of macrophages in the Fibrosis Pigeon breeders’ lung of SD rats suggesting that it is possible to change the direction of the polarization of macrophages by inhibiting Notch signaling pathway to affect the the Fibrosis Pigeon breeders’ lung of SD rats,providing a new therapeutic target for the clinic and influencing the development ofthe Fibrosis Pigeon breeders’ lung of SD rats. |
