Objective:To investigate effects of electro-acupuncture to the expression of oocytequality and apoptosis-related proteins Bcl-2,Bax in granulosa cells of kidney deficiencysenile infertile patients undergoing IVF-ET.Methods:Sixty-six senile infertile patients with tubal factors who were to undergoIVF-ET, were randomly assigned to two groups in which thirty-three cases in the treatmentgroup and thirty-three cases in the control group.To observe the differences ofimprovement of kidney deficiency syndrome,the amount of gonadotropin and days,theendocrine on HCG day,the number of harvested oocytes,the rate of high quality oocyte, therate of fertilization,the rate of high quality embryo and clinical pregnancy rate.To detect thelevels of Bcl-2and Bax proteins in granulosa cells by Western blot.Results:1.Compared with the control group,the kidney deficiency syndrome wasobviously better improved in the treatment group than that in the controlgroup(P<0.01).2.Compared with the control group,the treatment group in the amount ofgonadotropin and days,the Progesterone on HCG day reduced significantly(P<0.05),the E2of single oocyte on HCG day,high quality oocyte rate,fertilization rate,high quality embryorate increased dramatically(P<0.05),but there were no significant difference in the totalnumber of retrieved oocytes and the ratio of clinical pregnancy between the treatment andcontrol group(P>0.05).3.Compared with the control group,the level of Bcl-2protein ingranulosa cells was higher in the treatment group,but the level of Bax protein was lower(P<0.01).4.There were positive correlations among Bcl-2protein level and the E2of singleoocyte on HCG day,the total number of retrieved oocytes,the ratio of high qualityoocyte,fertilization,high quality embryos(P<0.01),a negative correlation was seen among Bcl-2protein level and the Progesterone on HCG day(P<0.01).There were negativecorrelations among Bax protein level and the E2of single oocyte on HCG day,the totalnumber of retrieved oocytes,the ratio of high quality oocyte,fertilization,high qualityembryos(P<0.05),a positive correlation was seen among Bax protein level and theProgesterone on HCG day(P<0.01).There was negative correlation between Bcl-2proteinand Bax protein levels(P<0.01).5.Compared with the non-pregnant patients,pregnantpatients showed significantly higher Bcl-2protein level and lower Bax protein level(P<0.05).Conclusions:1.Electro-acupuncture could obviously increase Bcl-2protein level andreduse Bax protein,inhibit granule cell apoptosis,improve ovarian function.2.There werecorrelations among Bcl-2and Bax protein levels and parameters of IVF.They could beused as important indicators of predicting developmental ability of the oocyte andembryo.3.Electro-acupuncture could improve the kidney deficiency syndrome,oocytequality and embryo quality of kidney deficiency senile infertile women.Its mechanismmight be associated with adjusting the expression of apoptosis-related proteins.
