Effect Of Mieyou Decoction On HSP70and AQP4in The Helico-bacter Pylori Associated Gastritis Mice Of Spleen-stomach Damp-heat Syndrome

Objective:To observe the expression of HSP70and AQP4in the mouse model of helicobacter pylori associated gastritis with spleen-stomach damp-heat syndrome and explore the possible mechanism of Mieyou decoction in treating helicobacter pylori associated gastritis with spleen-stomach damp-heat syndrome.Methods:(1)70BABL/c mice were divided into control group, model group, high concentration Mieyou decoction group,low concentration Mieyou decoction group,Gastric triad group. The mice of control group were given normal feeding; the model group, high concentration Mieyou decoction group, low concentration Mieyou decoction group,Gastric triad group:Using the composite factors (high-sugar and high-fat diet, hot and humid environment, helicobacter pylori infection) to make the mouse model of helicobacter pylori associated gastritis with spleen-stomach damp-heat syndrome; after the success of modeling,continuously intragastric administration for14days: the rest groups were given the corresponding drugs respectively.(2)Index detection:Using HE staining methods under the light microscope to observe the pathological changes of gastric mucosa of mice;using the western blot;immunohistochemistry to detect the expression of HSP70protein and AQP4protein in gastric mucosa.Results:The general situation:the physical condition of high concentration Mieyou decoction group, low concentration Mieyou decoction group, gastric triad group were improved; and the high concentration Mieyou decoction group were better than gastric triad group.The expression of HSP70:compared with control group, the expression of HSP70in BALB/c mice model group was significantly increased (P<0.01); compared with the model group, the HSP70in high concentration of mieyou soup group, low concentrations of mieyou soup group, gastric triad group were increased,the differences were statistically significant (P<0.01); The expression of HSP70in gastric triad group and high concentration mieyou soup were increased, the difference was not statistically significant(P>0.05).The expression of AQP4:compared with control group, the expression of AQP4in the model group was significantly increased (P<0.01); compared with the model group, the AQP4in high concentration of mieyou soup group, low concentrations of mieyou soup group, gastric triad group were increased,the differences were statistically significant (P<0.01); The expression of AQP4in gastric triad group and high concentration mieyou soup were decreased, the difference was not statistically significant(P>0.05).Conclusion:Mieyou decoction is better than Gastric triad in improving the general physical condition of the mice; Mieyou decoction may play a role in treatment of helicobacter pylori associated gastritis with spleen-stomach damp-heat syndrome by eradicating the helicobacter pylori, reducing the inflammation of the BALB/c mice, raising the expression of HSP70, reducing the expression of the AQP4, promoting the repair of the gastric mucosa.
