Object This study was designed to explore the possible mechanism of Danqi Baoxin particles treating CHF by observing the general situation,cardiac ultrasound,cardiac function and tumor necrosis factor-α,interleukin-6, brain natriuretic peptide of Chronic heart failure model in rats.Method One hundred male and female Sprague-Dawley(SD) rats were selected as research subjects, and10of them were randomized into blank group, the others(n=90) were established by intraperitoneal injection of adriamycin as CHF model group. After the model was built, using echocardiography to measure left ventricular ejection fractions(LVEF),left ventricular fractional shortening(LVFS),left ventricular internal dimension diastole (LVIDd) and left ventricular internal dimension systole (LVIDs),etc. After the model was established successfully,The CHF model group rats were randomly divided into Danqi Baoxin particles high-dose group,middle-dose group,low-dose group,Enalapril group and none-dose model group. Each treatment group was gavaged with high-dose、middle-dose、low-dose Danqi Baoxin particles and Enalapril for30days. Normal group and none-dose model group were gavaged with water for30days. The cardiac ultrasound and cardiac function of each group were measured by echocardiography, BNP, TNF-a and IL-6were measured by Elisa. Then apply statistical analysis to explore the effect of Danqi Baoxin particles to each indicator.Result1、 After the model establishment,Compared with the blank group, EF、FS of the CHF model group decreased obviously, LVIDd, LVIDs, EDV, ESV increased significantly,the difference was statistically (P<0.05).2、The end of the experiment,compared with the model group, EF、FS of Danqi Baoxin particles each dose group increased significantly, LVIDd,LVIDs,EDV,ESV decreased, the difference was statistically (P<0.05).3、The end of the experiment,compared with the model group, BNP of Danqi Baoxin particles each dose group were obviously decreased,the difference was statistically (P <0.05).4、The end of the experiment,compared with the model group, the levels of TNF-α IL-6in Danqi Baoxin particles each dose group were reduced significantly,the difference was statistically (P<0.05).5、The pathologic results showed:compared with model group, drug intervention groups of myocardial cell mild edema, inflammatory infiltration significantly reduced, especially in Danqi Baoxin particles high-dose group can obviously improve the adriamycin induced myocardial injury, has obvious therapeutic effect.Conclusion1、CHF animal model can be built successfully by intraperitoneal injection of adriamycin;2、Danqi Baoxin particles can improve cardiac function, reduce the level of BNP, improve cardiac ejection fraction and left ventricular short diameter shortening rate;3、Danqi Baoxin particles can alleviate myocardial inflammation and reduce the secretion of TNF-α and IL-6;4、Danqi Baoxin particles can inhibit the occurrence of ventricular remodeling, preventing myocardial cell hypertrophy and myocardial fibrosis and slowing down the process of CHF.
