Objective:to observe the effect of medical ozone water on rabbit knee osteoarthritis and its mechanisms; to observe the effect of ozone and nedical ozone water on rabbit knee osteoarthritis’cartilage and inflammatory mediators IL-1β、TNF-α and MMP-13 to explore which is more suitable for the treatment of knee osteoarthritis in rabbits; to observe :he effect of medical ozone water on rabbit’s cartilage and inflammatory mediators IL-1β、TNF-α and MMP-13.Methods:20 male and 20 female New Zealand white rabbits were randomly divided into five groups. These groups were blank group(group A), medical ozone water safety group(group B), model group(group C), ozone group(group D) and medical ozone water group(group E). Each group included 8 rabbits. In group A, the rabbits were fed normally. In group B, the rabbits were treated by medical ozone water once a week. In group C、 D and E, the rabbits were built into models by injection of papain. After having been moulded successfully, the rabbits in group C were fed normally without doing other managements, the rabbits in group D were treated by ozone and the rabbits in group E were treated by medical ozone water once a week. After three weeks’treatment, all the rabbits in each group were executed by the way of ear vein injection of air. The cartilage specimens were decalcified, embedded in paraffin, sliced, HE staining made into pathology. IL-1β、TNF-α and MMP-13 was detected using enzyme-linked immunosorbent assay(ELISA) method.Results:1.Compared with group C, the inflammatory mediators IL-1β、TNF-α and MMP-13 in group E decreased significantly (P<0.05).2.Compared with group D, the inflammatory mediators IL-1β、TNF-α and MMP-13 in group E decreased significantly (P<0.05).3.There is no significant difference between group A and group B on the evel of IL-1β、TNF-α and MMP-13 (P>0.05).Conclusion:Medical ozone water can inhibit the inflammatory mediators IL-1β、TNF-α and MMP-13 in rabbit knee osteoarthritis’cartilage, which suggests that medical ozone water can delay the progression of rabbit knee osteoarthritis. Compared with ozone, the inflammatory mediators IL-1β、 INF-α and MMP-13 in group treated with medical ozone water decreased significantly, which suggests that medical ozone water is more suitable for the treatment of knee osteoarthritis in rabbits. When medical ozone water is injected into normal rabbit knee joint, it will hardly cause damage on cartilage, compared with cartilage of normal rabbit knee joint, there is no significant difference.
