| Objective:1.To establish rabbit knee osteoarthritis animal models successfully, then to select optimal MR imaging sequences for their articular cartilages by grouping study2.To explore the affect of CT guided03intervention treatment of osteoarthritis in rabbits, to observe the expression of IL-1-ã€TNF-a and MMP-13on experimental rabbit articular cartilage3.To investigate the effect of ozone on the cartilage cells and its mechanism by comparing the immunohistochemical expression of articular cartilage of rabbit knee arthritis and the subtle changes in articular cartilage displayed on MRIMethods:1.The establishment of animal modelsEstablish the rabbit model of knee osteoarthritis by4%papain0.3ml intra-articular injection of rabbit knee joint two hind legs. 2. Animal groupsExperimental animals will be randomly divided into the normal group, the osteoarthritis group, the medical ozone group and the sodium hyaluronate group.The normal group and osteoarthritis group were left untreated. In the sodium hyaluronate group,1ml of SH with1%concentration was injected into bilateral knee cavity weekly for three times. Meanwhile, in the medical ozone group,5ml of ozone with35ug/ml concentration was injected into bilateral knee cavity weekly for three times too. MRI examination were carried out in bilateral knee joint of experimental animals just before killing them.3.To detect the expression of IL-1ã€TNF-a and MMP-13in the cartilage tissueTake articular cartilage of bilateral femoral medial condyle and cut into1×1×2mm3small strip. DAB method was used to detect the expression of IL-1ã€TNF-a and MMP-13in the cartilage tissue.Results:1.The rabbit knee osteoarthritis animal model were established successfully.2.Before and after the intervention of ozone treatment of rabbit knee osteoarthritis, variation of IL-1ã€TNF-a and MMP-13were found and their trends in the cartilage tissue were consistent.3.Positive expression rates of IL-1ã€TNF-a and MMP-13:The higher positive expression rates were found in the osteoarthritis group, sodium hyaluronate group and ozone group than that of in normal group,whereas the ozone group and sodium hyaluronate group were lower than osteoarthritis group. The difference between medical ozone group and the sodium hyaluronate group was not statistically significant.4.The results of MRI and immunohistochemistry showed that intra-articular injection of ozone or sodium hyaluronate for rabbit knee joints can retard the development of arthritis. There were no difference of statistically significant between the wo therapies.Conclusion:1.Intra-articular injection of ozone for rabbits with knee osteoarthritis can cause the expression changes of three kinds of inflammatory mediators:IL-1-. TNF-a and MMP-13. That were consistent with the immunohistochemical results.2.There were less IL-1ã€TNF-a and MMP-13in articular cartilage after the intra-articular injection of ozone for rabbits with knee osteoarthritis, that mean the injection may have rehabilitating effect for articular cartilage. Moreover those phenomenons of the cartilage as more smooth surface,less crakes, cells proliferation, etc appeared. So the reducing of those expression correspond to the pathological and MRI changes, could be used as evaluation index of treatment of OA. |
