Objective: To study the HMGB1(high mobility group box 1) and TLR4(Toll-like receptors 4) in ectopic endometrium, eutopic endometrium and normal endometrium expression and significance in the organization.Methods: using real-time fluorescence quantitative PCR and Western-blo tting detection of HMGB1 and TLR4 in 45 cases of patients with endometriosis, ectopic endometrium and eutopic endometrium and 30 cases of patients with non-endometriosis eutopic endometrium(normal control group) on the expression of m RNA and protein.Results:HMGB1, TLR4 m RNA in ectopic endometrium, eutopic endometrium and non-endometriosis eutopic endometrium(normal control) were expressed, ectopic endometrial groups of expression were the normal control group of 2.76 times, 2.68 times, respectively. the amount of expression of eutopic endometrium group as normal control group was 1.48 times, 1.56 times, respectively. and the comparison with the difference groups is significant(p<0.05), Western-blotting test results showed that the protein expression of HMGB1, TLR4 in ectopic endometrium, eutopic endometrium was higher comparing to the normal control group was low expression, and the results were consistent with real-time fluorescence quantitative PCR results.Conclusion: 1. HMGB1, TLR4 m RNA and protein were high expressed in endometriosis tissues, which maybe play an important role in the genesis, development in ovarian endometriosis. 2. HMGB1, TLR4 in the ectopic and eutopic membrane with Ems were higher than the expression of normal endometrium, show that endometrial inflammation may be the reason for the existence of the occurrence and development of Ems.
