With the improvement of people's living standards and the deepening of scientific research,some progress has been made in the prevention and treatment of malignant digestive system tumors,but the number of new cases and deaths of gastric cancer in China is still high.Gastric adenocarcinoma is the most common pathological type of gastric cancer.Its onset process is complex and long.It involves a variety of complex regulatory mechanisms,which are closely related to abnormal cell proliferation and abnormal cell apoptosis.In many malignant tumors,the abnormal proliferation of cells and the process of cell apoptosis are inhibited to varying degrees.If this pathological state persists,it will lead to the formation of tumors.Prostate apoptosis response protein-4(Par-4)is closely related to apoptosis.Its selective domain for apoptosis induction cancer cells(SAC)can promote apoptosis of cancer cells without affecting normal cells.At present,it is rarely reported in China that the expression of Par-4 in malignant tumors,especially in gastric adenocarcinoma.Cell cycle and apoptosis regulator 1(CCAR1)is a kind of perinuclear phosphorylated protein required in apoptotic signaling pathway.It plays an important role in regulating cell proliferation and apoptotic signal by combining with corresponding cell cycle regulator proteins and apoptotic regulator proteins,and also has abnormal expression in some malignant tumors.There are few studies on CCAR1 expression in malignant tumors,and there is no report on the expression of CCAR1 in gastric adenocarcinoma.Western blotting,immunohistochemical SP and qRT-PCR have not been reported to study the expression of Par-4 and CCAR1 in gastric adenocarcinoma and the correlation between them.In this study,the expression of Par-4 and CCAR1 in gastric adenocarcinoma tissues and adjacent normal gastric mucosa tissues was studied by immunohistochemical SP method,Western blotting and quantitative real-time polymerase chain reaction(qRT-PCR).The relationship between the expression of Par-4 and CCAR1 in gastric adenocarcinoma tissues and clinicopathological features was also explored.Whether there is a certain correlation,discuss their role in the occurrence and progression of gastric adenocarcinoma,so as to provide more accurate and efficient molecular biological indicators for patients in the early diagnosis and prognosis of gastric adenocarcinoma,which may enrich the molecular diagnosis and treatment targets further of gastric cancer.ObjectiveTo study the expression of Par-4 and CCAR1 in gastric adenocarcinoma and normal gastric mucosa adjacent to cancer,and to explore the relationship between the expression of Par-4 and CCAR1 and the clinicopathological features of gastric adenocarcinoma.Methods1 Immunohistochemical SP method and Western blotting was used to detect the expression levels of Par-4 protein and CCAR1 protein in gastric adenocarcinoma tissues and normal gastric mucosa adjacent to cancer.2 The expression levels of Par-4 mRNA and CCAR1 mRNA in gastric adenocarcinoma tissues and normal gastric mucosa adjacent to cancer were detected by qRT-PCR.Results1 Immunohistochemical results showed that the positive expression rates of Par-4 protein and CCAR1 protein were 28.3% and 81.6% in gastric adenocarcinoma tissues,82.5% and 32.5% in normal gastric mucosa tissues adjacent to cancer,respectively.Compared with normal gastric mucosa adjacent to cancer,the positive expression rates and intensity of Par-4 protein in gastric adenocarcinoma decreased,while the positive expression rates of CCAR1 protein were 82.5% and 32.5% in normal gastric mucosa adjacent to cancer.And the positive expression intensity increased,the difference was statistically significant(P<0.05,P<0.05).1.1 In gastric adenocarcinoma tissues,the expression of Par-4 protein was not related to the sex and age of patients,but had no significant difference(P all>0.05).The expression of Par-4 protein was related to lymph node metastasis,TNM stage,differentiation degree and infiltration depth of gastric adenocarcinoma(P all<0.05).The expression of CCAR1 protein was not correlated with sex,age,TNM stage,depth of invasion and lymph node metastasis of gastric adenocarcinoma,there was no significant difference(P all>0.05).But was correlated with the differentiation of gastric adenocarcinoma(P<0.05).1.2 There was no significant correlation between the expression of Par-4 protein and CCAR1 protein in gastric adenocarcinoma(P>0.05).2 Western blotting assay showed that the relative expression of Par-4 protein in gastric adenocarcinoma tissue was significantly lower than that in normal gastric mucosa adjacent to cancer,and the relative expression of CCAR1 protein was significantly increased(P<0.05,P<0.05).2.1 In gastric adenocarcinoma tissues,the expression of Par-4 protein was not related to the sex and age of patients,but had no significant difference(P all>0.05).The expression of Par-4 protein was related to lymph node metastasis,TNM stage,differentiation degree and infiltration depth of gastric adenocarcinoma(P all<0.05).The expression of CCAR1 protein was not correlated with sex,age,TNM stage,depth of invasion and lymph node metastasis of gastric adenocarcinoma,there was no significant difference(P all>0.05).But was correlated with the differentiation of gastric adenocarcinoma(P<0.05).2.2 There was no significant correlation between the expression of Par-4 protein and CCAR1 protein in gastric adenocarcinoma(P>0.05).3 The results of qRT-PCR assay showed,compared with normal gastric mucosa adjacent to cancer,the relative expression of Par-4 mRNA in gastric adenocarcinoma tissues was significantly down-regulated,while the relative expression of CCAR1 mRNA was significantly up-regulated,the difference was statistically significant(P<0.05,P<0.05).3.1 In gastric adenocarcinoma tissues,the expression of Par-4 mRNA was not related to the sex and age of patients,but had no significant difference(P all>0.05).The expression of Par-4 mRNA was related to lymph node metastasis,TNM stage,differentiation degree and infiltration depth of gastric adenocarcinoma(P all<0.05).The expression of CCAR1 mRNA was not correlated with sex,age,TNM stage,depth of invasion and lymph node metastasis of gastric adenocarcinoma,there was no significant difference(P all>0.05).But was correlated with the differentiation of gastric adenocarcinoma(P<0.05).3.2 There was no significant correlation between Par-4 and CCAR1 in gastric adenocarcinoma tissues(P>0.05).Conclusion1 The expression of Par-4 protein and its mRNA is low in gastric adenocarcinoma,which is related to the differentiation degree,TNM clinical stage,invasion depth and lymph node metastasis of gastric adenocarcinoma,suggesting that Par-4 is involved in the occurrence and progression of gastric adenocarcinoma.2 CCAR1 protein and mRNA are highly expressed in gastric adenocarcinoma,but not related to TNM stage,depth of invasion and lymph node metastasis,suggesting that CCAR1 protein and mRNA are involved in tumorigenesis,but may not be related to the progress of gastric adenocarcinoma.3 At protein level and gene level,there was no significant correlation between the expression of Par-4 and CCAR1 in gastric adenocarcinoma tissues,indicating that there was neither antagonism nor synergism between them.
