1、The serum 25(OH) D levels influence YKL-40 and airway inflammation in a murine model after the intervention of vitamin D.Obejective: Observe serum 25(OH) D levels after supplementation of different doses of vitamin D, and its influence on the concentration of Ig E, YKL-40 and bronchoalveolar lavage fluid cell counts and changes in lung tissue, and to explore the role of vitamin D supplementation and mechanisms in a mouse model with asthma, as well as the appropriate serum 25(OH) D levels and understand the role of YKL-40 in asthma.Methods: 60 BALB / c mice which were 4 to 5 weeks old, female and clean were randomly divided into normal control group A, asthma control group B, asthma and 1000 IU / kg vitamin group C, asthma and 2000 IU / kg vitamin group D, asthma and 3000 IU / kg vitamin group E, asthma +and 4000 IU / kg vitamin group F. The different dose of vitamin D was fed through gavage a week before sensitization, group A and group B was controlled with NS. Mice were sensitized and challenged with ovalbumin, vitamin D was fed until the end of the experiment. ELISA was used to detect serum 25(OH) D, serum Ig E, serum and bronchoalveolar lavage fluid YKL-40 concentrations, BALF eosinophil count proportion by Wright-GMS, HE staining of lung tissue.Results: 1) Serum 25(OH) D levels: group A(65.69 ± 2.47) nmol / L, group B(46.11 ± 3.87) nmol / L, group C(85.27 ± 3.26) nmol / L, group D(103.02 ± 4.55) nmol / L, group E(122.45 ± 6.31) nmol / L, group F(137.46 ± 2.89) nmol / L, the concentration gradually increased with the increasing amount of supplementary.Serum 25(OH) D levels in group B was lower compared with group A, comparison between the groups was different(P <0.05). 2) BALF and serum concentrations of YKL-40: Both the BALF and serum concentrations of YKL-40 in group D are the lowest except group A. The concentrations in group F was significantly higher than in group B(P <0.05). The difference was statistically significant. No significant difference(P> 0.05) between group C and groups E. Compare the BALF and serum concentrations within group, there were no differences between group C and D(P>0.05). 3) Ig E concentration: in asthma groups, group D [(5.02 ± 0.19) ng / m L] was the lowest compared with other groups, including the control group [(4.05 ± 0.17) ng / m L](P<0.05), group C[(5.70 ± 0.27) ng / m L] and E [(5.79 ± 0.24) ng / m L], group B [(6.35 ± 0.33) ng / m L] and F [(6.43 ± 0.40) ng / m L] had insignificant difference(P> 0.05). 4) cell counts and EOS(%):The cell counts and EOS% in group D were significantly lower than other asthma group(P <0.05), group F were significantly higher than that in group B(P <0.05), no significant difference between group C and E(P > 0.05). 5) lung tissue: There was no obvious inflammatory cells in group A.. Group B and Group F showed increased bronchial mucosal folds,a large amount of inflammatory cells infiltration around the lumen,the smooth muscle and basement membrane thickening,lumen stenosis,and a lot of secretions intraluminal, which was more evident in Group F. In group C and E, bronchus spasm mild, epithelial cells and smooth muscle hyperplasia mild, slightly thickened basement membrane, less peribronchial inflammatory cell infiltration. In group D, no obvious epithelial cells and smooth muscle proliferation, no obvious wall thickening, less inflammatory cell infiltration, but if compared with group A, mentioned above were slightly increased. 6) BALF YKL-40 and serum Ig E level were positively correlated(r=0.834,P<0.01), BALF YKL-40 level and EOS% of BALF were positively correlated(r=0.910, P<0.01), the difference was statistically significant.Conclusions: 1) Compared with the control group, serum 25(OH) D levels in asthma were lower,with the increase in vitamin D supplementation, serum 25(OH) D levels could be increased. 2) YKL-40 may play an important role in the pathogenesis of asthma, the concentration of YKL-40 may be a mark of the severity of asthma, and had a positive correlation with Ig E and EOS% level. 3) vitamin D supplementation in asthmatic mice can produce a dual role, the proper amount of vitamin D supplementation(2000IU / kg) may improve lung inflammation in mice, reduce the secretion of Ig E, YKL-40 and eosinophil infiltration, but the excess will increase the inflammatory response. 4) in mice with asthma, the appropriate serum 25(OH) D levels which can effect on immune system may be around 100nmol/L, if above 120-130nmol/L, it could aggravate airway inflammation. 5) Vitamin D may directly or indirectly affect the secretion of YKL-40, YKL-40 may also influence the absorption and action of vitamin D.2、the impact of different duration of sunshine on serum 25(OH) D levels in miceObejective: To understand the basic serum 25(OH) D levels in mice, observe the effects of different duration of sunshine on 25(OH) D levels, discuss the possible reason for insufficient of Vitamin D.Methods: 24 BALB / c mice which were 4 to 5 weeks old, female and clean were randomly divided into group A(control group), group B(sunlight <0.5h), group C(0.5h <sunlight <1h), group D(sunlight> 1h). The environment and diet had no differences between groups, the mice were exposed to sunlight directly in autumn for 6 weeks. ELISA was used to detect serum 25(OH) D concentrations.Results: Serum 25(OH) D levels of each group :group A(65.37 ± 2.80) nmol/L, group B(65.40 ± 2.84) nmol/L, group C(67.59 ± 1.93) nmol/L, group D(79.02 ± 1.74) nmol/L, there were significant differences between group D and the other three groups(P <0.01), the difference between group A, B and C was not statistically significant(P> 0.05).Conclusions: 1) The deficiency of Vitamin D is common at present, and is closely related with asthma. 2) The supplementation of Vitamin D and the duration of sunshine need further study.
