ObjectiveTo explore the impact of herbal compound Huangqin decoction on effects of related factors on RORγt protein, Interleukin-17(IL-17) and Interleukin-23(IL-23), and its possible advantage.MethodsThe 40 Sprague-Dawley(SD) rats in four groups at random: the normal control group, the asthma model group, the budesonide(BUD) group, the huangqin decoction group, 10 rats in each group. The model of asthma was established by OVA sensitizing and challenging. Drug intervention for the first time after the excitation. After six weeks, the count and classification of the inflammatory cells in peripheral blood were detected. Detect the counts and classification of inflammatory cells in blood, The pathological characters of lung tissues were discovered by using hematoxylin and eosin(HE) staining. The level of IL-17 in serum and BALF were determinated by enzyme-linked immunosorbent assay(ELISA). The levels of IL-23 protein in pulmonary tissue were detected by immunohistoche-mistry. The expression of RORγt protein in pulmonary tissue were measured by Western blot.ResultsCompared with the normal group, the airway inflammation were obviously Increased in the asthma group, the BUD group and the Huangqin decoction group. Compared the BUD group and the Huangqin decoction group with the asthma group, the airway inflammation were significantly reduced. Compared the BUD group with the Huangqin decoction group, there were no significant diffirence in the airway inflammation.Compared with the normal group, the count and classification of the inflammatory cells in blood were obviously Increased in the asthma group, the BUD group and the Huangqin decoction group(P<0.05). Compared the BUD group and the Huangqin decoction group with the asthma group, the count and classification of the inflammatory cells in blood were significantly reduced(P<0.05). Compared the BUD group with the Huangqin decoction group, there were no significant diffirence in the above indexes(P>0.05).Compared with the normal group, the content of IL-17 in serum and BALF and the level of IL-23,RORγt protein in pulmonary tissue were obviously Increased in the asthma group, the BUD group and the Huangqin decoction group(P<0.05). Compared the BUD group and the Huangqin decoction group with the asthma group, the content of IL-17 in serum and BALF and the level of IL-23,RORγt protein in pulmonary tissue were significantly reduced(P<0.05). Compared the BUD group with the Huangqin decoction group, there were no significant diffirence in the above indexes(P>0.05).ConclusionsThe Huangqin decoction can reduce asthmatic rats lung bronchioles cell infiltration by blocking inflammation chain reaction. The mechanism which the Huangqin decoction inhibits Th17 cells involved in asthma probably include decreasing the expression of IL-17, IL-23 and RORγt.
