Objective:To observe the B cell activating transcription factor (B-cell activiting transcription factor, BATF), retinoic acid-related orphan nuclear receptors (retinoic acid-related orphan recepptor γ t, ROR Y t), interleukin-17(IL-17) expression in asthmatic mice spleen tissue explore the BATF and its mechanism in the pathogenesis of asthma, looking for the new ways of treatment of asthma. Methods:15 healthy female BALB /c mice were randomly divided into normal control (NS) group, asthma group (AS), dexamethasone (DXM) group, each group of mice are 5. AS group, DXM mice at days 0,7 and 14 with ovalbumin (OVA) 100 ug aluminum hydroxide (Al (OH) 3) 1mg mixing of saline suspension 0.2ml intraperitoneal injection sensitization NS group with normal saline instead of intraperitoneal injection; 21 days NS group received 0.9% saline, AS group and DXM group to the 1% OVA ultrasonic atomizing inhalation, day 1, every 40 minutes, continuous 5 days excitation; the DXM group at 30 minutes before stimulation, dexamethasone l.Omg/kg intraperitoneal injection of NS group and AS group be the same amount of 0.9% saline by intraperitoneal injection same time. All mice were sacrificed 24 hours after the last challenge. Bronchoalveolar lavage fluid (BALF) line white blood cell count and differential count.FIE staining to assess the mouse airway and lung tissue inflammatory cell infiltration; enzyme-linked immunosorbent assay (ELISA) to detect the amount of IL-17 in spleen tissue; Real-time PCR method to detect BATF mRNA in spleen tissue, ROR γ t mRNA Expression level. Results:(1) lung histopathology HE staining:AS were observed in small airways and small perivascular inflammatory cell infiltration, mucosa and submucosa edema, alveolar spaces visible eosinophils, neutrophils, monocytes cells and overflow of red blood cells; the DXM group of inflammatory cells, exudate than AS group, the basic integrity of the bronchial epithelium, and absorb visible signs of inflammation; the NS group no significant small airway mucosal edema, smooth lumen, without occlusion, around small blood vessels not See the infiltration of inflammatory cells. (2)Compared with NS group, the total number of leukocytes in BALF of the AS group, the number of neutrophils and eosinophils increased significantly (P<0.05), DXM in BALF total number of leukocytes, neutrophils and eosinophilsnumber also increased (P<0.05), compared with the AS group, DXM in BALF total number of leukocytes, neutrophils, and eosinophils was significantly lower (P<0.05). (3) spleen homogenates of IL-17 level:the AS group IL-17 the highest level of DXM group followed by the lowest NS group, the difference between the three groups was statistically significant (P<0.05), and compared with NS group AS group and DXM group IL-17 water were significantly increased, and the difference was statistically significant (P<0.05), DXM group compared with the AS group to reduce the level of IL-17, and the difference was statistically significant (P<0.05). (4) in the spleen tissues BATF mRNA ROR γ t mRNA expression levels:the highest level of AS group, DXM group followed by the NS group was the lowest, the difference between the three groups was statistically significant (P<0.05), and compared with NS group, AS group and DXM group BATF mRNA ROR Y t mRNA expression was significantly increased, the difference was statistically significant (P<0.05), DXM group compared with the AS group BATF mRNA ROR γ t mRNA expression level of the difference was statistically significant (P<0.05). (5) The correlation analysis showed that:Spleen tissues of the IL-17 concentration in BALF WBC count, neutrophil count and eosinophils count positively correlated (r= 0.838,0.737,0.882, P<0.005).BATF mRNA expression of ROR γ t mRNA expression of IL-17 level was a significant positive correlation (r = 0.807,0.833, P<0.005). The spleen homogenates IL-17 concentration and spleen tissues RORyt mRNA expression levels were positively correlated (r= 0.890, P<0.005).Conclusion:(1) BATF and RORyt regulation of IL-17 secretion, involved in the development of airway inflammation and proinflammatory role of IL-17 (2) dexamethasone may by BATF and RORyt partly be able to prevent the spleen-derived Th17cell differentiation, reduce the secretion of IL-17, thereby improving cell infiltration of airway inflammation, relieve asthma symptoms.
