| Objective To investigate the therapeutic effect of Jiawei Huangqin Decoction on ulcerative colitis and the regulation of Signal transducer and activator of transcription 3/Nuclear factor kappa-B/Interleukin-6 pathway.Methods 48 mouses were randomly divided into blank group,model group,positive drug group(Salicylazosulfapyridine),low-dose group,middle-dose group and high-dose group with 8 mouses in each group.According to 3%DSS moulding method,ulcerative colitis was induced in 5 groups of mouses except blank group.After 7 days,the blank group and the model group were given saline by gavage,while the drug treatment group was given corresponding drugs by gavage.The gavage volume was 1 Oml/kg body weight for a week.After treatment,mouses were killed by cervical dislocation and colon length was measured.The changes of colon histomorphology and histopathological score were observed by HE staining.The expression levels of STAT 3,NF-kB,IL-6 in colon tissues of mouses in each group were detected by RT-qPCR and Western blot.Results Compared with the blank group,the colon length of mice in the model group,the positive drug group and the Chinese medicine group was significantly shorter(P<0.05),and the pathological score of colon tissue was significantly higher(P<0.05);compared with the model group,the colon length of mice in the positive drug group and the Chinese medicine group was significantly longer(P<0.05),and the histopathological evaluation of colon tissue was made.Compared with the positive drug group,the colon length of mice in the high-dose group was significantly longer(P<0.05),and the colon histopathological score was significantly lower(P<0.05).There was no significant difference in colon length and colon histopathological score between the middle-dose group and the low-dose group(P>0.05).Compared with the high dose group,the colon length of mice in the middle and low dose group was significantly shortened(P<0.05),and the colon histopathological score was significantly increased(P<0.05);compared with the high dose group,the colon length of mice in the low dose group was significantly shortened(P<0.05),and the colon histopathological score of mice in the low dose group was significantly increased(P<0.05).Length was significantly shortened(P<0.05),and colon histopathological score was significantly increased(P<0.05).Compared with the blank group,the expression levels of STAT 3,NF-kB,IL-6 in the colon tissue of the model group were significantly higher(P<0.01).Compared with the model group,the expression levels of STAT 3,NF-kB,IL-6 in the colon tissue of the positive drug group and each dose group of traditional Chinese medicine were significantly lower(P<0.01),and the expression levels of STAT 3,IL-6 in the colon tissue of the high dose group of traditional Chinese medicine were significantly lower(P<0.01).The levels of expression of NF-kB and IL-6 were significantly lower than those of the positive drug group and the middle and low dose groups of Chinese medicine(P<0.05),while the levels of STAT 3,NF-kB and IL-6 in colon tissue of the middle dose group had no significant difference from those of the positive drug group(P>0.05),but were significantly lower than those of the low dose group of Chinese medicine(P<0.05).Conclusion Jiawei Huangqin Decoction can improve ulcerative colitis,and its mechanism may be related to down-regulating the expression of STAT 3,NF-kB and IL-6 in colonic tissue through affecting STAT 3/NF-kB/IL-6 pathway. |
