Objective: 238 differential expression proteins related with gastric cancer were found by quantitation proteomics research in our prophase research. We selected the Cartilage oligomeric matrix protein(COMP) and heat shock protein 90α/β(HSP90α/β) proteins of the more differential multiple and observed the expression of both in gastric cancer tissue, then analyzed the clinical significance of both expression.The results will provide valuable experiment datas for gastric cancer patients on clinical early diagnosis, clinical pathology grouping and evaluation the clinical prognosis. Methods: The 8 cases samples include of normal gastric mucosa, cancer side tissue and gastric cancer were respectively collected and extracted the total protein of these tissues. The expression of COMP and HSP90α/β proteins was analyzed by Western-blot in normal gastric mucosa, cancer side tissue and gastric cancer tissues. Then, the expression of two proteins was observed by immunohistochemistry staining in gastric tissue arrays(included of normal gastric mucosa, cancer side tissue and gastric cancer). Last, the clinical pathology significance of both expression was analyzed in gastric cancer tissue.Results: 1. The Western-blot results displayed that the expressions of COMP and HSP90α/βproteins were higher in gastric cancer tissue than that in normal gastric mucosa and cancer side tissue(P<0.05). Both expressions were higher in cancer side tissue than that in normal gastric mucosa(P<0.05).2. The immunohistostaining results showed that:(1) the expression positive rates of COMP protein were respectively 58.93%(33/56) and 56%(14/25) in male and female patient gastric cancer tissues. The expression positive rates of COMP protein were respectively 5.88%(2/34), 26.92%(7/26) and 58.02%(47/81) in normal gastric mucosa, cancer side tissue and gastric cancer tissue. The expression positive rates of COMP protein were respectively 40.00%(2/5), 52.94%(9/17) and 61.01%(36/59) in gastric well differentiated, middle differentiated and poorly differentiated adenocarcinoma. The expression positive rates of COMP protein were respectively 57.14%(28/49) and 59.38%(19/32) in the tumor volume ﹥3.0cm and ≤3.0cm gastric tissues. The expression positive rates of COMP protein were respectively 46.15%(12/26) and 70.91%*(39/55) in no lymphaden metastatic and lymphaden metastatic tissues. The expression positive rates of COMP protein were respectively 46.88%(15/32) and 73.47%(36/49) in Ⅰ-Ⅱperiod and Ⅲ-Ⅳperiod patient gastric cancer tissues no lymphaden metastatic and lymphaden metastatic tissues.(2) The expression positive rates of HSP90α/β protein were respectively 53.57%(30/56) and 52%(13/25) in male and female patient gastric cancer tissues. The expression positive rates of HSP90α/β protein were respectively 11.76%(4/34), 30.77%(8/26) and 65.43%(53/81) in normal gastric mucosa, cancer side tissue and gastric cancer tissue. The expression positive rates of HSP90α/β protein were respectively 40.00%(2/5), 58.82%(10/17) and 69.49%(41/59) in gastric well differentiated, middle differentiated and poorly differentiated adenocarcinoma. The expression positive rates of HSP90α/β protein were respectively 65.31%(32/49) and 65.66%(21/32) in the tumor volume ﹥3.0cm and ≤3.0cm gastric tissues. The expression positive rates of HSP90α/β protein were respectively 42.31%(11/26) and 69.09%(38/55) in no lymphaden metastatic and lymphaden metastatic tissues. The expression positive rates of HSP90α/β protein were respectively 56.25%(18/32) and 79.59%(39/49) in Ⅰ-Ⅱperiod and Ⅲ-Ⅳperiod patient gastric cancer tissues.3.The expressin positive rate of COMP and HSP90α/β proteins was higher in gastric cancer tissue than that in normal gastric mucosa and cancer side tissue(P<0.05). Both expression positive rate was higher in cancer side tissue than that in normal gastric mucosa(P<0.05). Both expression positive rate was higher in lymphaden metastatic gastric cancer tissue than that in no lymphaden metastatic gastric cancer tissue(P<0.05). Both expression positive rate was higher inⅢ-Ⅳperiod patient gastric cancer tissue than that in Ⅰ-Ⅱperiod(P<0.05).Conclusion: The expression of COMP and HSP90α/β proteins were related with gastric cancer effluence, differentaiation level of gastric cancer tissue, lymphaden metastasis and TNM classification of gastric cancer. As the differentaiation level of gastric cancer tissue decreased, lymphaden metastasis of gastric cancer and TNM classification increased, the expression rate of two proteins was also heightened.
