Expression And Clinical Significance Of Pim-1and RON Proteins In The Gastric Cardia Cancer

Objective:The incidence of the gastric cardia cancer shows an increasing trend. Unfortunately, the natural course of the gastric cardia cancer is long and the clinical symptom is concealing. Many patients are discovered in the advanced stage. Patients have a poor prognosis due to tumor invasion and metastasis. The occurrence and development of the gastric cardia cancer is related to a variety of factors. However, the molecular biological mechanisms are not yet fully elucidated. The study is used to detect expression levels of Pim-1proto-oncogene protein and RON protein in gastric cardia cancer tissues, cardia intestinal metaplasia tissues and normal cardiac tissues by immunohistochemistry and to analyze relations of Pim-1and RON proteins with clinical pathological characters. The role of Pim-1and RON proteins in the development and prognosis of the gastric cardia cancer and the correlation between the expression of Pim-1and RON in the gastric cardia cancer is further explored, which provides some theoretical basis to clinical diagnosis, treatment and prognosis.Methods:A total of95cases of the gastric cardia tissue were collected, containing56cases of surgical specimens of the gastric cardia cancer,21cases of biopsy specimens of the cardia intestinal metaplasia,18cases of biopsy specimens of the normal cardia. Expression levels of Pim-1and RON proteins in56cases of gastric cardia cancer tissues,21cases of cardia intestinal metaplasia tissues and18cases of normal cardiac tissues were detected using immunohistochemistry. We respectively chose the positive histological section as positive control of Pim-1and RON proteins with PBS replacing the antibody as negative control. Brown particles were observed in the cytoplasm and nucleus as the positive reaction of Pim-1and in the cytoplasm as the positive reaction of RON. The staining results depended on the staining intensity and the percentage of the positive cell. SPSS13.0software was applied for all analysis and statistical significance was defined as P values less than0.05.Results:(1)Expressions of Pim-1protein were mainly localized in the cytoplasm, with a few located in the nucleus. RON protein was expressed in the cytoplasm. Both of them showed brown particles from weak to strong.(2)The positive expression rates of Pim-1and RON proteins in gastric cardia cancer tissues, cardia intestinal metaplasia tissues and normal cardiac tissues were78.6%,38.1%,5.6%and66.1%,33.3%,11.1%, respectively. There was a statistically difference in positive expression rates of Pim-1and RON among different gastric cardia tissues (P<0.05).(3)The positive expression rate of Pim-1in T4was statistically higher than that in T1~T3(P<0.05). The positive expression rate of Pim-1in the group without lymph node metastasis was statistically lower than that in the group with lymph node metastasis (P<0.05). There was no statistically difference in the positive expression rates of Pim-1among different gender, age, tumor differentiation, TNM stage (P>0.05).(4)The positive expression rate of RON in T4was statistically higher than that in T1~T3(P<0.05). The positive expression rate of RON in the group without lymph node metastasis was statistically lower than that in the group with lymph node metastasis (P<0.05). The positive expression rate of RON in stage Ⅲ~Ⅳ was statistically higher than that in stage Ⅰ~Ⅱ (P<0.05). There was no statistically difference in the positive expression rates of RON among different gender, age, tumor differentiation(P>0.05).(5)There was no correlation between the expression of Pim-1and RON in the gastric cardia cancer (P>0.05).Conclusion:(1)Expression levels of Pim-1and RON in gastric cardia cancer tissues, cardia intestinal metaplasia tissues and normal cardiac tissues show an increasing trend, which suggests that Pim-1and RON may play a role in the occurrence and development of the gastric cardia cancer. They may have some clinical value to the diagnosis, treatment.(2)The expression of Pim-1in both the cytoplasm and the nucleus may participate in the genesis of the gastric cardia cancer.(3)In respect of the relation between Pim-1and RON proteins and clinical pathological characters, their expressions may be associated with tumor invasion, metastasis. They are expected to be new molecular markers to diagnose and monitor the gastric cardia cancer and evaluate patients’prognosis.(4)There may be no significant or direct correlation between Pim-1and RON proteins in the development of the gastric cardia cancer.
