ObjectiveCell autophagy is to meet the environment pressure,formation in cells with double layer or multilayer coating wrap package organelles and devour bubble formation,thus the degradation and transformation of organelles,end in cell apoptosis.A recent study found that abnormal autophagy function of many tumor cells,but its mechanism is still unclear,and the molecular mechanism of autophagy and the relationship between autophagy and cancer for cancer prevention and control has great significance.astric cancer is the world within the scope of one of the highest morbidity and mortality of mali-gnant tumor,its first in a number of digestive system malignant tumor in our country.This study proposed by immunohistochemical detection of autoph-agy related protein Beclinl,P62 and IDH1expression in gastric cancer and tissue adjacent to carcinoma.And analyze the experimental data,study the correlation between the three and cancer of the stomach and the relationship between the clinical pathological features.And then discusses the change of autophagy activity and gastric cancer occurrence,development,regulatory mechanism for autophagy provide reliable theoretical basis for research in gastric cancer.MethodsThis study collected during October 2012-October 2012(the people's liberation army eighty-eight hospital)gastric cancer surgery,postoperative pathology experts confirmed for64 cases of gastric cancer tissue samples tissue adjacent to carcinoma in 64 cases(from 2cm tumor tissue and outside the organization).By immune histochemical method(S-P method)tag with IDH1 Beclin1,P62 protein expression in gastric cancer tissue and tissue adja-cent to carcinoma,and combining with the patient's age,sex,tumor size,degree of differentiation,whether with lymph node metastasis,TNM staging and infiltration depth of clinical pathology data analyzed.All experimental data by SPSS 20.0 statistical software to complete the processing of statistics and count data by chi-square test methods compared between groups;The Spearman correlation analysis,the correlation between variables inspection and consistency with the Kappa test analysis;Level data using rank and inspe-ction(the Mann-Whitney test)compared between groups.Alpha=0.05 for the inspection level.This study collected during October 2012-October 2012(the people's liberation army eighty-eight immune histochemical method(S-P method)to detect Beclin1 protein,P62 protein,IDH1 protein expression in gastric cancer tissue and tissue adjacent to carcinoma cases,analysis of Beclin1 protein and protein P62,IDH1 activity of protein expression and tumor tissue pathological type(stage),infiltration depth,and presence of lymph node metastasis and other indicators of relevance,and combine all the clinical pathologic data statistical processing for the data from experiment.Results1.Beclin1 in gastric cancer cell membranes and are expressed in the cytoplasm,for tan color,strong positive nuclei,mesenchymal cells also dyeing.P62 protein in gastric cancer tissues in the positive expression mainly in the nucleus.Isocitrate dehydrogenase 1(IDH1)the positive expression of protein aggregation in cell plasma.Beclin1,P62 and IDH1 in gastric cancer tissue of the positive expression rate was 53.1%(34/64),46.9%(30/64),54.7%(35/64).In the tissue adjacent to carcinoma positive expression rate was50.0%(32/64),28.1%(18/64),84.3%(54/64).Three kinds of protein expre-ssion level difference in different organizations by the rank and inspection,all have(z_s=3.599,P<0.001)(z_s=2.350,P=2.350),(z_s=5.149,P<0.001).2.Beclin1 in high,medium and low differentiation of gastric cancer tissues positive expression rate was 58.3%(7/12),69.2%(18/26),34.6%(9/26),P62 in high,medium and low differentiation of gastric cancer tissues positive expression rate was 66.7%(8/12),57.7%(15/26),26.9%(7/26),IDH1 in high,medium and low differentiation of gastric cancer tissues positive expression rate was 33.3%(4/12),46.2%(12/26),73.1%(19/26),their differences were statistically significant(P<0.05).P62 is related to tumor diameter of5 cm or more(chi-square=20.127,P<0.001);P62,IDH1 is related to whether lymph node metastasis(chi-square=10.458,P<0.001),(chi-square=10.092,P=0.001).3.The Spearman correlation analysis results show that Beclin1 P62 protein expression with no obvious correlation,(r_s=0.192,P=0.192;Kappa=0.191,P=0.124).Beclin1 was positively correlated with IDH1 protein expre-ssion(r_s=0.403,P<0.001;Kappa=0.403,P<0.001).IDH1 and negatively corr-elated with P62 protein expression(r_s=0.277,P=0.027;Kappa=0.274,P=0.274).Conclusions1.Beclin1,IDH1 protein expression in gastric cancer tissues,p62 in gastric cancer tissues expression level rises,prompt autophagy levels in gastric cancer tissue.2.Beclin1 protein expression level and the degree of the gastric cancer tissue differentiation,lymph node metastasis,invasion depth,tumor diameter of 5 cm or more;IDH1 expression level and tissue differentiation,lymph node metastasis of gastric cancer and tumor diameter of 5 cm or more;P62 protein expression level and the degree of the gastric cancer tissue different-iation,lymph node metastasis and tumor diameter of 5 cm or more.3.Beclin1 in(3)in gastric cancer and the expression of IDH1 correlates,p62 and negatively correlated with the expression of IDHI,Beclin1 and p62 express no correlation,suggest we may have a certain role in gastric cancer occurs in three.
