Objective:In this study,acupuncture combined with music therapy was used to treat anxiety disordered rats. By exploring changes in behavior and T lymphocytes.It was expected to explore the effect of acupuncture combined with music therapy in anxiety disordered(AD) rats,and it was also looked forward to explicating the role of the therapy in immunologic mechanism of anxiety disorder. To provide new ideas for anxiety disorder research.Methods: Female SD rats were randomly divided into groups of normal(N), model(M), acupuncture therapy(AT),music therapy(MT) and acupuncture combined with music therapy(AMT).Chronic unpredictable emotional stress lasted 21 days to establish the AD rat model following with Light on or off for 24 h, overcrowding, pooling the rats from two cages into one for 24 h,isolation by placing a single rat into one cage for 24 h,new hierarchy by mixing 4 rats from two different cages for 24 h, tilting the cages for 24 h,limited access to food or water for 24 h,inescapable foot shock(20 shocks,duration 5s,intensity 10 m A,interval 30s)The sequence of individual stress exposures was the same in all rats except the normal group.All the rats were observed with weight changes during 21 days.On day 7,14 and21, behavioral activity was measured using the elevated plus-maze and open-field test in all rats.Then messured the spleen index and thymus index. Observing T lymphocytes subgroup by using flow cytometry.Results1.Changes in weight gainBody weight gain in rats showed different changes.The results showed thatcompared with the N,weight gain in each group was significantly decreased(P <0.01);compared with the M,weight gain in AT and AMT was significantly higher(P<0.01).2.Behavior tests2.1Elevated Plus-maze TestIn addition to AMT no significant difference in OT% and OE% on 21d(P> 0.05),OT% and OE% in all the other groups were significantly lower than N(P <0.01, P <0.05); OT%,OE% in AT,MT and AMT were significantly higher than M on 21d(P <0.0l, P <0.05);OT% in AMT were significantly higher than MT on 21d(P <0.0l).2.2Open-field TestHorizontal scores and Vertical scores in M,AT,MT and AMT were significantly lower than N at each time point(P<0.01);horizontal scores in AT,MT,AMT were significantly higher than M on 14 d to 21d(P<0.01).Horizontal scores in ATM were much higher than AT on 21d(P<0.01).Vertical scores in AT,MT and ATM were significantly higher than M on 21d(P<0.01).Vertical scores in ATM were much higher than MT and AT on 21d(P<0.05, P<0.01). 3.Spleen index and Thymus index.Data showed that thymus index and spleen index of rats in each group had no significant difference(P> 0.05).4. T lymphocytes subgroupThe data showed CD8~+ in M were significantly lower than N(P<0.01), CD4~+/CD8~+ in M were significantly higher than N(P<0 05);CD8~+ in AT and AMT were significantly higher than M(P<0 01).CD4~+/CD8~+ in AT and AMT were significantly lower than M(P<0 05).Conclusion1.Regulation to behavior of anxiety disorder model rats in AT,MT and AMT, suggesting that the three therapy has a good anti-anxiety effect.AMT was most effective.2.CD8 was significantly lower in M, CD4/CD8 was significantly higher in M. ATM,AM could increase CD8~+ and decrease CD4~+/CD8~+ in M.It was suggested thatATM and AT could regulate the level of T lymphocytes in anxiety disorder rats. |
