Male Wistar rats were randomly allocated to five groups as follows: normal control, AD model control, AD model plus EA, AD model plus music, AD model plus both EA and music. To observe the behavioral changes of model rats through Elevated Plus Maze Test System (EPM) and record the frequency of open-arms entries (OE)and the duration of open-arms time(OT) in anxiety model rats. The concentration of 5-HT in rat hypothalamus and plasma were determined by HPLC-ECD.Results 1.Compared to normal group, the OE% and OT% of model group are both decreased. It showed that we successfully build the anticipant AD model. 2. EA or music or EA & music tended to increase OE% and OT%. 3. The 5-HT in hypothalamus of AD model rats was significantly higher than that in normal group. On the contrary, the 5-HT in plasma of AD model rats was definitely lower than that in normal group. The significantly lower contents of 5-HT had been observed in hypothalamus of AD model rats treated by either EA or music. Moreover, the same result had been gotten as well by using them together. To play music could decrease 5-HT in plasma of model rats. However, to stimulate the point or give both EA and music cannot obtain the same result.Conclusion The three different treatments can decrease the anxiety level of AD model rats. It is suggested that EA or appreciate music or EA & music may have potential of anti-anxiety by regulating the higher level of 5-HT in hypothalamus of AD model rats.
