Objective: This study was to observe the results of the form and proliferation of mouse fibroblast cells conducted by serum containing Jiangzhuo particles Chinese pharmaceutics by pharmacological method. We obtained serum containing the Jiangzhuo particles by intragastric administration of normal rabbits. According to the results, we could discuss and study the prevention and mechanism of action to renal fibrosis by Jiangzhuo particles Chinese pharmaceutics.Methods: Screened 16 healthy male normal rabbits and randomly divided into four groups(4 / group), and conducted intragastric administration according to the experiment design after normal fed for a week and obtained required serum with Jiangzhuo particles. Intragastric administration taken rabbits were fed pharmaceutics for a week, once a day. Fasted for 12 hours but the water.① Control group: Intragastric administration by normal saline, 10 ml / L ? d; ② Low-concentration group: Intragastric administration by low-concentration of Jiangzhuo granular compound preparations. Effective dose of Jiangzhuo granular compound preparation is 2.1g /( kg ? d); ③Medium-concentration group: Intragastric administration by medium-concentration of Jiangzhuo granular compound preparations. Effective dose of Jiangzhuo granular compound preparation is 4.2g /(kg ? d); ④ High-concentration group: Intragastric administration by medium-concentration of Jiangzhuo granular compound preparations. Effective dose of Jiangzhuo granular compound preparation is 8.4g /(kg ? d). Prepared the desired serum with compound preparations after the last administration for 1h by arteria carotis blood taken. Normal mouse fibroblast cells were cultured in the medium. Medium were replaced the medium once a day.The fibroblast cells were subcultured once every three days. Collected the log phase of mouse fibroblast cells with serum-free DMEMmedium, made of 2x106 / ml suspension and inoculated to 6 well plate. Every amount of cell suspension per well was 2.5ml. After 18 h, culture medium was replaced and started the experiment.Results: In this study, three different concentrations of serum containing compound preparations were added to fibroblast cells and cultured. The results showed that the inhibition level to the proliferation of mouse fibroblast cells is different with the concentrations of the serum containing compound preparations. The G0 / G1 phase of three serum containing Jiangzhuo granule groups consisted longer time in cell cycle than the control group. However, the S phase of three serum containing Jiangzhuo granule groups consisted shorter time in cell cycle than the control group. According to the above results, the G0 / G1 phase and the S phase demonstrated a dose-dependent relation to the concentrations of serum containing compound preparations.The results had significantly statistical difference(P <0.05).Conclusion: The high, medium and low concentrations of serum containing Jiangzhuo particles could inhibit the growth and mitosis of fibroblast cells and also induce apoptosis of fibroblast cells. The inhibition effect of serum containing Jiangzhuo granular compound preparations to the proliferation of vitro mouse fibroblast cells increased with the concentrations of serum.
