Objective: To intervene the burned rats with different concentrations of hydrogen sulfide,to seperate and detecte the percentage of fibroblasts by flow cytometry,to discuss the effect of exogenous H2 S on burned rats skin fibroblasts proliferation. Method:Adopt fibroblasts from burned rats as study object. To select 90 healt hy SD rats and randomly divide into 15 groups(normal control group on 3rd d ay, 7th day, 14 th day, normal saline group on 3rd day, 7th day, 14 th day, Na HS group on 3rd day, 7th day, 14 th day, PPG group on 3rd day, 7th day, 14 th day, NaHS+PPG group on 3rd day, 7th day, 14 th day), The normal control group rec eived no intervention, the rats in rest of the groups were created 5% TBSA p artial thickness burn wound, the rats in normal saline group were timely initiat ed intraperitoneal injection of saline daily, the rats in Na HS group were timely initiated intraperitoneal injection of Na HS solution daily, the rats in PGG gro up were timely initiated intraperitoneal injection of propargyl glycine solution d aily, the rats in NaHS+PGG group were timely initiated intraperitoneal injectio n of NaHS + propargyl glycine solution daily, to separate fibroblasts from the wound on the 3rd, 7th and 14 th day after burn; fixed and stained the fibroblas t to detect changes of cell cycle by flow cytometry, and were analyzed using one-way ANOVA, test level take α=0.05.Results: At the same time point, to compare the percentage of fibroblasts in S phase, there were significant difference between normal control group and nor mal saline group(P<0.05); the percentage of fibroblast in S phase of normal s aline group were lower than the NaHS group(N1, N2, N3), the difference was statistically significant(P<0.05); the percentage of fibroblasts in S phase of N aHS group(N1, N2, N3) were higher than the Na HS+PPG group(NP1, NP2, N P3), the difference was statistically significant(P<0.05); the percentage of fibro blasts in S phase of NaHS+PPG group(NP1, NP2, NP3) were higher than PPG group(P1, P2, P3), the difference was statistically significant(P<0.05); the percentage of fibroblasts in S phase of PPG group(P1, P2, P3) were lower than t he normal saline group, the difference was statistically significant(P<0.05). Conclusion: The fibroblasts could be proliferated after body burn. Low dose of exogenous hydrogen sulfide could promote burn wound fibroblast proliferation; PPG is the inhibitor of H2 S synthase, by inhibition of endogenous H2 S generation can be suppressed burn wounds fibroblast cell proliferation. H2 S has the role of promote the burn wound healing.
