Objective:To investigate the effect of huaiqihuang cream(HQH)on proliferation,cell cycle distribution and the expression of p21,Cyclin A in mouse mesangial cells(MMC).Methods:1.The mouse mesangial cells were divided into 4 groups: normal control group and HQH treatment group(8 mg/ml,16 mg/ml,and 24 mg/ml).2.Use cck-8 assay to detect the proliferation of mesangial cells after treated by HQH for 24 h,48 h and 72 h.Calculate the inhibition rate and compared with the normal control group.3.Use flow cytometry to detect the cell cycle distribution of mesangial cells after 48 h treatment with HQH,and compared with the normal control group.4.After 48 hours of treatment with HQH,use western blotting to detect the expression of Cyclin A and p21 proteins in each group.Results:1.Observed by inverted microscope,the cell morphology after HQH treatment is different from that of normal control group.2.After 24,48,and 72 hours,the absorbance of each HQH group was significantly lower than that of the control group,suggesting that the above concentration of HQH has a blocking effect on the proliferation of mouse mesangial cells(P <0.01).3.After 48 hours,the results of flow cytometry showed that the proportion of cells inS phase was significantly higher(P <0.05),indicating that HQH may blocking cell cycle in S phase.4.After 48 hours,the expression of cell cycle-related proteins CyclinA and p21 was detected by western blotting.The results showed that the expression of CyclinA was significantly down-regulated,while the expression of p21 was significantly up-regulated(P <0.01).Conclusion:A certain concentration of HQH has inhibitory effect on the proliferation of mouse mesangial cells,and this effect may be related to its effect on the expression of cell cycle-associated proteins,and thus the cell cycle is arrested in the S phase.
