Research background According the latest research report which is released by WHO and International Agency for Research on Cancer (IARC), affected by the environment、diet、lifestyle and the aging of the population,the burden added by new cancer in developmenting country is out of proportion with the burden in the world.There are more than sixty percent of cancer cases in Africa, Asia and Latin America. at the same time, because of the lack of timeliness of cancer early screening and treatment, the area had taken over 70% of cancer deaths, colorectal cancer is one of the three highest rate of cancer worldwide popular, there are at least 400000 patients died from colorectal cancer each year. China as a developing country, since reform and opening up, has the incidence of colorectal cancer increased gradually. Prognosis is different in colorectal cancer early compared with late, so a large-scale screening is very significant. The studies showed, in nearly 20 years, the colorectal cancer is due to the changes of genetic material and the formation of abnormal gene caused by environmental, genetic, diet, lifestyle, drugs and medical aspects. UCPs is a transporter protein on the mitochondrial inner membrane, which can mediate proton leak, and reduce the generation of ATP and reactive oxygen species. So far, it has discovered 5 homologs in mammals, namely UCP1, UCP2, UCP3, UCP4, UCP5.UCP2 is a kind of inner mitochondrial membrane anion transport protein found in 1997, which consists of 8 exons and 7 introns, molecular weight of 33218, consisted of 309 amino acid. In recent years, because of its wide distribution and various functions, People pay more and more attention on correlation between UCP2 and the biological behavior of malignant tumor.In order to investigate the effect of UCP2 in carcinogenesis, invasion and metastasis of colorectal cancer, this study investigates the expression of UCP2 in different colorectal mucosas by immunohistochemistry, and tries to analyze the formation mechanism of carcinogenesis and metastasis in colorectal cancer.Purpose To explore the role and clinical significance of UCP2 in development,infiltration and metastasis of colorectal adenocarcinoma.Methods All The cases were selected from general surgery and pathological department in my hospiytal between 2010,June and 2013,June.all patients were not receiving chemotherapy or radiotherapy before surgery,and a complete fowwoe-up data.including 20 cases of normal colorectal tissue,20 cases of colorectal adenoma,and 50 cases colorectal adenocarcinoma. the expression of UCP2 in normal colorectal tissue,colorectal adenoma and colorectal adenocarcinoma were detected by immunohistochemical analysis.Results The results of immunohistochemistry showed the difference of UCP2 proteins positive expression among the normal colorectal tissue,the colorectal adenoma and the colorectal adenocarcinoma were significant (P<0.05). the positive expression of UCP2 was not correlated with patient age、gender and degree of differentiation (P>0.05).the positive expression of UCP2 was closely correlated with depth of tumor infiltration、metastasis of lymph node and cancer embolus (P<0.05)Conclusion UCP2 may play an important role in tumorigenesis,infiltration and metastasis of colorectal adenocarcinoma.
